Patrmanová, Eliška ; Štveráková, Tereza (advisor) ; Křížová, Barbora (referee)
This diploma thesis is focused on the comparison and usability of the Short Form (SF) and the Complete Form (CF) of the test battery Bruininks-Oseretsky Test of Motor Proficiency, Second Edition (BOT-2) in individuals after the treatment for acute lymphoblastic leukemia (ALL). In the theoretical part, an overview of findings related to ALL, assessment of motor development, and especially the BOT-2 test battery is given. The main goal of the practical part was to verify the comparability of results from the most commonly used forms of BOT-2 in patients after ALL treatment. Methodology: 37 participants aged 4-21 years (mean age 10.3 years; SD ±3.9) were included in the study. The level of motor proficiency was measured with the BOT-2 CF. Data for the SF was extracted from the CF and processed using Two-Way Analysis of Variance (ANOVA), Receiver Operating Characteristic (ROC) analysis, and correlation analysis. Results: Two-Way ANOVA results showed that post-ALL patients achieved a significantly (p < 0.001; Hays ω2 = 0.41) lower standard score in the SF (45.1 ±7.9) compared to the CF (49.1 ±9.4). Acceptable sensitivity (72.3 %) and high specificity (91.9 %) of the BOT- 2 SF were proven. Conclusion: The BOT-2 SF can be used to screen overall motor proficiency and classify an individual's result in the...

Štveráková, Tereza ; Matoušková, Petra (advisor) ; Skarková, Veronika (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Kralove Department of Biochemical Sciences Author: Bc. Tereza Štveráková Supervisor: doc. Ing. Petra Matoušková, Ph.D. Consultant: Ing. Helena Kovaříková, Ph.D. Title of diploma thesis: Monitoring changes in microRNA expression in cervical cancer Cervical cancer is the fourth most common cancer among women globally. Prevention of cervical cancer is based on routine screening using quality diagnostic tests. This diploma thesis focused on searching for sensitive and specific diagnostic markers that would detect the early stage of carcinogenesis. Suitable candidates for a diagnostic markers are microRNAs (miRNAs), which negatively post-transcriptionally regulate gene expression. In carcinogenesis, miRNAs are deregulated and may play a significant role in cancer development. The aim of this diploma thesis was to identify significantly deregulated miRNAs that could be used as diagnostic markers of cervical cancer. In this diploma thesis, the combination of miR-423-5p and miR-25-3p was selected as the endogenous miRNA control based on the results of real-time PCR analysis and the results of the RefFinder algorithm. After that, the relative expression of selected miRNAs (miR-10b-5p, miR-145-5p a miR-182-5p) was determined by real-time PCR and...

The influence of balance aids to better quality of handwriting of chidren with ADHD
Holubová, Michaela ; Kobesová, Alena (advisor) ; Štveráková, Tereza (referee)
The children from all over the world suffer from ADHD (Attention Deficit Hyperactivity Disorder) and this disorder is a great social, educational and medical problem at schools. This bachelor thesis focuses on the etiopathogenesis, symptoms, diagnosis and treatment. The pharmacological treatment is not the only way which effectively helps children with their problems at school and in personal life. Exercise and regular physical activity is very important for children and it can impact on symptoms of ADHD. The experimental section of this bachelor thesis deals with the influence of balance aids to handwriting of children with ADHD. The thesis compares their handwriting before and after three months, during which the children were using the aids for the dynamic sitting. The experimental section includes four casuistics of children with ADHD, the results of standardized test MABC-2 for the evaluation of children's fine and gross motor skills and balance and a detailed analysis of writing from the point of view of specific features. The effect of therapy was evaluated using the total score which was recalculated to percentiles in the test MABC-2, handwriting was evaluated using specific features in the script. The therapy revealed itself as effective. Keywords ADHD, test MABC-2, sensory processing,...
Gross motor skills development and assessment in children aged 6-10 - a systematic review
Králová, Vilma ; Štveráková, Tereza (advisor) ; Nováková, Pavlína (referee)
This bachelor's thesis deals with the issue of gross motor skills development in children of younger school age (6-10) and with the possible options of its assessment. The theoretical part is devoted to definition of the term younger school age. It also defines important milestones in child's motor development and sets general recommendations about physical activity at this age. Tests evaluating gross motor skills have been described, both clinical tests - Timed Up and Go Test (TUG), Six Minute Walk Test (6MWT), Ten Meter Walk Test (10MWT), Time to Rise from the Floor (TRF), and test batteries Movement Assessment Battery for Children - 2nd Edition (MABC-2), Bruininks-Oseretsky Test of Motor Proficiency - 2nd Edition (BOT-2), MOBAK (Motorische Basiskompetenzen), Test of Gross Motor Development - 3rd edition (TGMD-3) and Körperkoordinationstest für Kinder (KTK). On a sample of four probands the gross motor skills have been measured using test batteries MABC-2, BOT-2 Short Form (SF) and MOBAK. The main aim of this study was to evaluate and compare subtests through all the test batteries which have been chosen. We did not find any significant correlation between results of MABC-2 and BOT-2 SF. However, the children's results in all the test batteries can be rated as average to above average. None of...

The diploma thesis invastigates the effect of specifically designed six-week clinical Pilates program on posture and low back pain. The theoretical part contains an overview of the Pilates method - the history, principles and its usage in rehabilitation. It also briefly describes the concept of Dynamic Neuromuscular Stabilization (DNS), which, along with the original Pilates method and the Spiraldynamic concept, has created the current Pilates Clinic Method. Furthermore, one chapter is devoted to the system of training Pilates teachers in the Czech Republic. The main objective of the practical part is to evaluate and compare the effect of an organized and an individual six-week Pilates exercise programme in patients with faulty posture and low back pain. Subsequently, constitutive objectives are set on the basis of selected examination methods. Firstly, evaluation of the exercise programme by the means of clinical objective tests combined into 5 groups: standing and balance, spinal mobility, DNS tests, somatognosia (shoulder width) and respiratory amplitude. Secondly, with the instrumental measurement of Bodystat's values: height, weight, waist circumference, hip circumference, fat, active body mass, water and body mass index. Thirdly, subjective evaluation using standardized questionnaires the...
