The preparation and the following in vitro saturation study of the radiopharmaceutical 99mTc-DTPA-ramucirumab on PC-3 cell line
Sabolová, Klaudia ; Bárta, Pavel (advisor) ; Janoušek, Jiří (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Student: Klaudia Sabolová Supervisor: Mgr. Pavel Bárta, Ph.D. Title of diploma thesis: The preparation and the following in vitro saturation study of the radiopharmaceutical 99m Tc-DTPA-ramucirumab on PC-3 cell line In cancer treatment, immunology is given prominence, which compared with chemotherapy and radiotherapy has a lower risk of side effects on healthy tissues. Immunotherapy includes application of monoclonal antibodies aimed at some tumour antigens using either non conjugated monoclonal antibodies or conjugated ones with an appropriate effector element, such as radionuclide. Angiogenesis plays the important role in pathogenesis of tumour diseases. Angiogenic process is regulated mostly by the interactions among vascular growth factors (VEGFs) and VEGF receptors (VEGFR). The main regulator of angiogenesis is VEGF-A. The blocking of the interaction among VEGF-A and its receptors VEGFR-1 and VEGFR-2 inhibits angiogenesis, and so does the growth of tumours. Ramucirumab is the monoclonal antibody with antiangiogenic effect, which blocks this interaction by its binding to the extracellular VEGFR-2 domain with high affinity. The presented study is focused on ramucirumab...
The study of properties of radiolabelled monoclonal antibodies for imaging in oncology
Janoušek, Jiří ; Trejtnar, František (advisor) ; Kozempel, Ján (referee) ; Lázníčková, Alice (referee)
Charles University, Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Candidate: Jiří Janoušek, MSc. Supervisor: Assoc. Prof. PharmDr. František Trejtnar, CSc. Consultant: Mgr. Pavel Bárta, Ph.D. Title of doctoral thesis: The study of properties of radiolabelled monoclonal antibodies for imaging in oncology Monoclonal antibodies belong to the group of targeted biological drugs that are currently successfully applied in various medical specialties, including oncology. The ability to specifically bind to antigens present in a tumour tissue is their significant attribute that can be used for imaging tumours in Nuclear Medicine. Receptors for vascular endothelial growth factor (VEGFR) and epidermal growth factor (EGFR) are possible tumour specific targets that are characteristic for some types of proliferating tumour tissue. The attachment of appropriate radionuclide is necessary for the potential radiodiagnostic use of monoclonal antibody. This task requires chemical modification of the antibody molecule that can damage its immunoreactivity and the binding to the target receptor. The aim of the doctoral thesis was to study the influence of labelling conditions with selected diagnostic radionuclides on radiochemical characteristics, in vitro receptor specific binding and...

Manufacturing of parts of microforming technologies
Janoušek, Jiří ; Peterková, Eva (referee) ; Podaný, Kamil (advisor)
This work deals with point incremental forming technology, which is now booming thanks to the possibility to program the exact path of a simple tool and to implement it on modern CNC machines. It solves the forces in the process, the roughness of the final surface of the products or their accuracy. This technology is divided into one-point and two-point forming and uses better material formability because of little local touch. Thanks to this, it is used in many fields such as medicine or the automotive industry. It is used to produce prototypes and specific components, which would be expensive.
Wireless charging station
Pokorný, Josef ; Hrabina,, Jan (referee) ; Janoušek, Jiří (advisor)
The paper deals with possibilities of wireless charging and transmission of electric energy using inductive coils. Basic models of air coils are described using CST simulation software. The product consists of a transducer for transmitting coil, two coils with ferromagnetic shield, synchronous rectifier and battery charging circuit. Communication is provided by an 8-bit microcontroller with wireless modules and an ethernet shield for online monitoring. The aim of the thesis is to design a wireless charging station with a power transfer of 40 W with the possibility of online monitoring of the charging state.
SPE Utilization in Analysis of Benzimidazoles I
Janoušek, Jiří ; Kubíček, Vladimír (advisor) ; Drastík, Martin (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Jiří Janoušek Supervisor: Ing. Vladimír Kubíček, CSc. Title of diploma thesis: Use of SPE in analysis of benzimidazoles I Anthelmintic resistance in parasitic worms is an up-to-date topic. In order to combat this resistance we need to find proper analytic methods of its monitoring. The goal of this thesis is to find a suitable method of benzimidazole extraction, using a solid-phase extraction. During the experiments I used SPE tubes Supelco DSC-C8 (1ml, 100mg). In my thesis I tested the influence of pH and composition of elution agents on recovery of extraction. I performed the extraction in acid, neutral and basic environment. Elution agents consisted of a mixture of acetonitrile and polar phase, with four different mixing ratios. The best results were obtained during extraction in basic environment. For SPE tubes DSC-C8 the most suitable elution agent consisted of 4 parts of acetonitrile and 6 parts of 10% ammonia solution. Keywords: benzimidazoles, flubendazole, mebendazole, solid-phase extraction, resistance, anthelmintics
Quarter-mode four-element antenna array
Janoušek, Jiří ; Fiala, Pavel (referee) ; Raida, Zbyněk (advisor)
Thesis deals with problematics of Compact Circularly Polarized Antenna Based on Quarter-Mode Substrate Integrated Waveguide Sub-Array and process. It also solves a complex structure of microstrip power distribution network. The goal was to create a~physical functioning model of this antenna and to measure its attributes in free space and also near the human body. Theoretical part of this work is taking on the calculation of parameters of the proposed antenna. Practical part is then solving modelation and simulations in program ANSYS HFSS. Physical model of the antenna was not created and some flaws were found in the presented model of the microstrip power distribution network. A possible solutions were proposed to eliminate these flaws.
Increasing the safety of the outside perimeter
Janoušek, Jiří ; Fiala, Pavel (referee) ; Marcoň, Petr (advisor)
This thesis deals with increasing the safety of the external perimeter. The theoretical part describes the parameters of areas exhibiting an increased safety risk. For these spaces, relevant safety hazards and corresponding elimination methods are determined. Then, the term "drone" is analyzed in the context of its applicability. The practical section characterizes the process of building and configuring unmanned aircraft to guard the perimeter and the designing and fabrication of an automatic charging station for the aircraft. The final part of the thesis is a brief manual describing correct handling of the created system.
