Substitute family care in the Czech Republic

This dissertation is concerned with the issues surrounding substitute family care in the Czech Republic and presents an analysis of its legal regulation as set out in both private law and public law regulations. The term substitute family care encompasses several independent legal institutions, which together form a system serving to protect children at risk who cannot grow up in their own family. The system of substitute care is divided into family-type individual care on the one hand and collective care provided in institutional facilities on the other. This dissertation examines the historical development of forms of substitute care, analyses their current legal regulation, including procedural regulation, and evaluates the interconnectedness and mutual harmoniousness of individual legal regulations. It analyses the inner workings of the substitute care system and delineates the rights and obligations of entities which enter into it in various capacities, with an emphasis on how the fulfilment of the rights of a minor is enforced and how their best interest is considered. Mentoring of ex-offenders as an example of social innovation in the Czech Republic

The concept of social innovation is increasingly becoming a subject of interest particularly due to the challenges associated with the crisis of the welfare state. The high rate of ex- offending is an example of a current challenge and mentoring is a social innovation which seeks to take up this challenge. The main goal of this thesis is to interrogate the process of social innovation using mentoring of ex-offenders as an example and to explore mentoring and its implementation within the Czech Republic. The theory of the process of social innovation, as documented in the overseas literature, guided the extensive research conducted in Czech non-governmental organizations (NGOs) that provide mentoring approach. The empirical part of this thesis provides a valuable insight into the process of social innovation from NGO perspective. It includes a comprehensive description of developmental stages of the process as well as highlights the limitations that NGOs encounter in implementing mentoring in the Czech Republic.
Novel Approaches in Electrochemical Determination of Xenobiotic Compounds and in Study of Their Interaction with DNA

Presented Ph.D. Thesis is focused on the development of analytical methods applicable for determination of selected xenobiotic compounds and for monitoring DNA damage they can induce. The main attention has been paid to the development and testing of non-toxic electrode materials for preparation of miniaturized electrochemical devices and novel electrochemical DNA biosensors. 2-Aminofluoren-9-one (2-AFN) was selected as a model environmental pollutant, which belongs to the group of hazardous genotoxic substances. Its carcinogenic and mutagenic effects may represent a risk to living and working environment. 2-AFN has one oxo group, where the cathodic reduction occurs, and one amino group, where the anodic oxidation occurs. The voltammetric behavior of 2-AFN in the negative potential region was investigated at a mercury meniscus modified silver solid amalgam electrode (m-AgSAE) representing a non-toxic and more mechanically robust alternative to mercury electrodes. This working electrode was subsequently used for the development of a newly designed miniaturized electrode system (MES), which has many benefits as the possibility of simple field measurements, easy portability, and the measurement in sample volume 100 µL. Title: A FOUNDATION PLAN OF KARATE SPORT CLUB Aims: The aim is to explain how to establish a karate sport club in new terms of foundation of associations. Another goal is the application of the theoretical knowledges to a chosen karate club Red Dragon Team (RDT) and to design options how to improve its functioning. Methods: Based on expert literature and texts research, a study has been produced which describes basic terms and ways of analysis execution. The SWOT analysis of the current state of the inner and outer environment of the RDT sports clubwas used in the second section of this document. Results: The work provides comprehensive guide describing the particular steps for founding of a new association in practice. The work is proposing a solution based on the SWOT analysis to eliminate possible deficiencies and how to better use the potential of the club RDT. Title: The popularity of the sport among the general public in relations to its presentation on TV Objectives: The aim of this thesis is to determine the popularity of the sport among the general public and to compare it with its presentation on TV. The popularity and viewership of the sport is determined by questioning the public. To conduct interviews with experts on sports broadcasters from television, explaining views of sport news provided on public and commercial televisions. Final results were compared with the results of observation presentation of the sport in sport news in the thesis of Trnková (2014). The results allowed to propose recommendations which will be for use in marketing, sports and media organisations. The results will only imply a certain degree of scientific contribution. Methods: The market research of this thesis has consisted of quantitative and qualitative methods. An electronic questionnaire published on the server Vyplňto.cz, helped gather results from the public. Interviews were carried out in-depth including electronic questioning via e-mail correspondence. Presentation of sports in television broadcasting in the Czech Republic

Title: Presentation of sports in television broadcasting in the Czech Republic Objectives: The aim of this thesis is to measure and compare the airtime devoted to individual sports and sports representations on public and commercial television in the regular, prime-time sports news. The recommendations will be proposed to be used in sports-club marketing, based on the results of observations. Methods: This thesis uses, as a method of observation, the standard sport news shown at prime-time on the TV stations ČT1 and TV Nova. This is the type of indirect observation with the technical devices - USB TV tuner and PC. In terms of content, Berelson's quantitative content analysis will be used, which examines the content of media messages. The theoretical part of the thesis is based on an analysis of documents. Results: The results of observations illustratively show the different approaches between public and commercial television for the inclusion of sport in the broadcasting of regular sports news in prime-time. For both channels, the most popular sports in the sports news are football and ice hockey. In addition, there is a trend of popularity of summer sports, even in the winter time. Title of diploma thesis: Modification of Glutathione S-transferase by selected glycating agents Glycation is a non-enzymatic reaction, which leads to the binding of carbonyl compounds to free amino groups of amino acids, peptides, proteins or other biomolecules. This process is closely related to impaired protein function in diabetes mellitus, during aging, and in neurodegenerative diseases. The main purpose of this thesis was to monitor the impact of several glycating agents on the properties of the enzyme glutathione S-transeferase (GST, EC 2.5.1.18) which plays a key role in the detoxification of many exogenous as well as endogenous compounds. Methylglyoxal (0.5-2 mM), glyceraldehyde (0.5-10 mM), glucose (50 mM), and fructose (50 mM) were selected as glycating agents. The bachelor´s thesis is focused on job-law relationships, specifically on system of employee rewarding for the selected employer. Target of the thesis is especially to optimize personal outlay of employer. Theoretical part is focused on clarification of relationships between employer and employees. Next part is devoted to the analysis of present state of rewarding of employee and questionnaire survey. Based on the analysis are suggested optimal solutions leading to economical and tax handiness.

The thesis is focused on analysis of physical activity of secondary students in Ceske Budejovice. The literature review describes the period of adolescence with its psychical and physical changes and deals with physical activity in this developmental stage. The research part is aimed at monitoring of physical activity of the students by pedimeter. Third-year students participated the research and monitored their physical activity through INDARES web portal. The results were processed statistically and presented in charts.

Plant polyphenolic compounds naturally occurring in human diet possess a wide range of biological and pharmacological properties. They can interact with various proteins including enzymes. Their interaction with serum albumin has a great significance because of its ability to bind, transport and store many endogenous and exogenous low-molecular compounds present in blood circulation. One of the objectives of this thesis was to study chemical structure-binding affinity relationships of two important groups of polyphenolic substances, namely hydroxycinnamic acids and catechins (flavanols), with the molecule of serum albumin in the in vitro models using spectroscopic (UV-Vis absorption spectroscopy, fluorescence quenching method) and electrophoretic (native and SDS PAGE) methods.
