Properties of aminoclay complexes and biologically active substances

This paper builds on previous research of aminoclay complexes in undergraduate studies. Theoretical part deals with study of current problems of aminoclay complexes with bioactive substances and the choice of substances for complexing with aminoclay. The experimental part consists of preparation of aminoclay complexes with selected bioactive agents at various concentrations. Verification of binding of bioactive agents to the aminoclay matrix was performed by Elemental Analysis (EA) and Fourier-transform infrared spectroscopy (FT-IR). For finding of the bound amount of bioactive substance were used the Ultra Performance Liquid Chromatography (UPLC®) and analysis of the cytotoxic properties of the formed complexes by used by the MTT assay. The main motivation of this study is to create new complexes with improved characteristics that would replace existing forms of substances used in pharmaceutical and biomedical applications.
Reflexes of laryngeals in the Akkadian loanwords from the West-Semitic Languages in the 1st millenium BCE

The aim of this work is to determine the patterns used by the speakers of Akkadian to adapt laryngeal phonemes in loanwords from West-Semitic languages by analysing the reflexes of these phonemes in the Akkadian words. Corpus used for analysis is taken mainly from already existing lists of West-Semitic loanwords in Akkadian (von Soden 1966, 1968, 1977; Cherry 2017; Zadok 2021, 2022) subsequently reduced using criteria based on Abraham - Sokoloff (2011) and supplemented by additional lexemes (mainly from CAD). The results of this analysis are further corroborated by analysing transcriptions of West-Semitic personal names occurring in RINAP 1 - 5 containing laryngeals into cuneiform. Findings of these analyses are then presented in the final part of the work. It was possible to determine quite unambiguously the individual patterns of adaptation of the respective laryngeal phonemes. Original /ḥ/ and /ḫ/ were adapted as /ḫ/, original /ˤ/ and /ˀ/ as /ˀ/ or ∅, depending on its position. Original /h/ and /ġ/ were not present in the studied corpus, thus precluding any possibility of determining their adaptation patterns. Development of the circadian system after systemic potentiation of GABAergic transmission in early ontogenesis

The circadian system is the evolutionary response of organisms to repetitive geophysical conditions and allows organisms to devote their energy to solving sudden changes. Although the circadian system developed to help organisms survive and thrive in dynamic conditions, system itself is sensitive to external changes. During critical developmental period of the mammalian CNS, the neonatal period, the circadian systém is significantly vulnerable. Benzodiazepines, pharmaceuticals widely prescribed for half a century, interfere with GABAergic transmission, which is critical type of signaling for the functioning of the circadian systém. The purpose of this thesis is to determine whether and how the development and functioning of the circadian system is altered after administration of a drug from the benzodiazepine class, clonazepam, during the critical developmental period. This thesis examines changes in the expression profile of selected genes in the hippocampus, olfactory bulb and frontal cortex of two ontogenetic stages of rats. Results of this work suggest that benzodiazepines administred in the neonatal period disrupt the development of the circadian system and that these changes, particularly in the hippocampus, persist for certain time. Shift of the comfort zone and personal development of participants in teambuilding events

This work deals with the shift of the comfort zone and personal development of participants in teambuilding events. The first part defines key terms such as teambuilding, comfort zone, personal development, motivation and stimulation. Next part presents survey using structured interviews. Respondents were asked about their experiences with the shift of the comfort zone and personal development at such events. The following part analyses the interviews to find out whether these teambuilding events have any impact on the personal development and shift of the participants' comfort zone.
Vyhodnocení růstu a zdravotního stavu telat v závislosti na různých chovatelských podmínkách

The goal of every livestock farmer should be a viable, well-growing healthy calf that passes on its genetic information to future generations. This thesis deals with the impact of growth and health of calves depending on the selected breeding technology in chosen farm of Holstein cattle. The aim was to compare various ways of housing calves in operating conditions. The chosen hypothesis predicts to achieve better results of calves housed in an individual system (VIB) compared to a group system (VSB). In 2020 a group of calves (n 41), divided evenly into various housing systems (individual /group), was monitored in the selected farm. The experiment lasted from the birth to the 85th day. Based on the results of the experiment, the calves of both observed groups received quality colostrum (average value of 26.17 % Brix) in the sufficient amount (average amount at the first drink 3.45 l). Calves are not weighed at birth in this farm (calves are assigned a value of 30 kg, regardless of sex). The average measured birth weight was 38.4 kg. It would be good to adjust this value in the in-house system to reflect reality better. On the basis of the results of observations, the original hypothesis can be rejected. There was no statistically significant difference between the two groups of calves when compared. It is not possible to confirm / exclude the degree of susceptibility of individual groups of calves to infectious pressures and diseases objectively (statistically) due to a small number of observations. Diarrheal diseases were more serious problem. These diseases occurred most in calves up to one month of their age, with a disease prevalence of 25 % and an average mortality of 7,4 %. No statistically significant difference was found when weight gains were compared. Weaned calves of the VIB housing system showed higher profitability, on average CZK 313 per calf, CZK 2.92 per kg of weight gain. From the point of view of animal welfare, there were problems with scraped backs of the calves housed in plastic sheds (Calf - Tel) because of their height at the shoulder. These sheds had a low entryway (95 cm). In the farm, the milk fed calves are fed from free surface; the teats are not used here. It would be worth considering and testing whether a change of a calf feeding method had a positive effect on the prevalence of diarrheal diseases. This could lead not only to a reduction in costs of potential veterinary care and medicine, but also to an increase in weight gain and thus to higher economic yields.
Optimalizace procesu nasazení technologie WebRTC

The diploma thesis deals with the optimization of deployment and infrastructure of systems based on WebRTC technology for streaming video and audio over the internet network, in which this technology enables creating P2P connections between endpoints. The thesis describes problems related to P2P access and finds an alternative in the form of a specific SFU server. The provided client application implements P2P communication method, which is compared with SFU thanks to the implementation of its own measuring apparatus and implements automated switching of both modes in the background of the ongoing conference and proposes further optimization changes for a continuous conference in order to reduce endpoint load.

Jan Dušek Supervisor Prof. PharmDr. Petr Pávek, Ph.D. Title of Doctoral Thesis Development of novel Constitutive androstane receptor (CAR) ligands Constitutive androstane receptor is nowadays known as the established nuclear receptor that regulates the expression of several key cytochrome P450 enzymes, predominantly CYP3A4 and CYP2B6. Recently it has been shown that CAR has also essential role in the regulation of endogenous metabolism of glucose, lipids, cholesterole or bile acids. Simultaneously, this receptor is considered to have proliferative effect on human hepatocytes and protective effects against toxic and dietary damage of liver parenchyme. Given the possible therapeutic utilization of CAR, its therapeutic options are being intensively studied. Unfortunately, currently known ligands of human or mouse CAR are either poorly selective or indirect. The aim of my doctoral thesis was to find new ligands of mouse and human CAR, that would enable more detailed study of the receptor.

The thesis includes a market research around commercially available timing devices, their comparison with each other according to available parameters, and finally a system is designed and constructed that combines the capabilities of already manufactured sets with new features and possibilities for large-scale extension beyond the boundaries of fire sport.
