Metalothioneiny u drobných volně žijících savců jako bioindikátor znečištění životního prostředí

Metalothioneiny (MT) jsou cysteinem bohaté nízkomolekulární proteiny, schopné vázat esenciální i toxické kovy. Jejich syntéza je indukována přítomností zejména kadmia a zinku, ale také dalších kovů jako Pb, Ni, Hg, Ag a i jinými faktory (např. stres, kortikoidy) 1. Pro práce věnující se studiu biologických funkcí metalothioneinů jako homeostáze, prevence oxidačního stresu, detoxifikace či studie vlivu znečištění životního prostředí na různé živočichy je důležitý celkový obsah metalothioneinů v jednotlivých tkáních 2. U vodních organismů, zejména Mytilus galloprovincialis 3 a Ruditapes decussatus 4 je obsah MT užíván jako bioindikátor znečištění mořské vody a je proto snaha mít obdobnou možnost pro zjišťování kontaminace půdy a životního prostředí.
Voltammetry of Metallothioneins on Mercury Meniscus Modified Silver Solid Amalgam Electrode

Peak of AgMT is formed on m-AgSAE from MT in alkaline media. The height of this peak is proportional to the MT concentration in range 37- to 301 ng /ml and can be increased with accumulation. In ammonia solution with Co(III) salt, apart from usual Brdička response at -1400 mV another, more positive catalytic system is observed, when formation of AgMT is allowed. This phenomenon has been confirmed by elimination voltammetry.
Determination of Metallothioneins in the Liver of Small Terrestrial Mammals, Living at Hazardous Element Contaminated Sites

Animals living in the contaminated areas are exposed to elevated concentrations of risk elements. Pfibramsko is one of most contaminated sites in Czech Republic. This area is loaded with sources of geological and metallurgical industry influence resulting in extremely high Pb, Cd, and Zn contents in soil. We analyzed eighteen subjects trapped on selected locations around Kovohute Pribram, belonging to the species Apodemus sylvaticus and Microtus arvalis. By differential pulse voltammetry and modified Brdicka reaction were measured concentrations metallothionein (MT) in their livers. Differences of concentrations between measured MT contents were found between species and among places of trapping.
The Use of Self-Assembled Monolayer of Thiolated Calix[4]arene on Polycrystaline Gold Electrode Surface

This study deals with the use of thiolated calixarene (C[4]A) as gold electrode modifier, alone or in combination with insulating undecanthiol (C11SH), which is used to fill the gaps between calixes. Desorption potential of thiolated C[4]A and C11SH are both pH dependent, cathodic desorption peaks can be recorded in basic solutions only. The properties of the modified electrode were investigated using hydroquinone - electrochemically active molecule which fits inside the C[4]A cavity. Obtained data suggest that the electrochemical oxidation/reduction ofhydroquinone does not use C[4]A cavities as size-selective channels.
Isolation and Characterization of Protoplasts and their Utilization for Model Membrane

Protoplasts are prepared form plant cells by removing their cell walls, which are usually destroyed mechanically or by enzymes. Protoplasts exhibit the spherical shape after removing of their cell walls. They can be isolated from a range of plant tissues: leaves, stems, roots, anthers, and even pollen. Plasmatic membranes surrounding protoplasts can be used as a source of model membranes. Therefore, some parts of plasmatic membranes or subcellular organelle membranes, which contain specific transport systems, can be isolated and integrated to model phospholipid membranes. State of such model membranes can be characterized by electrochemical impedance spectrometry (EIS) and the transported species can be determined with the use of voltammetric methods.
Transport of Phytochelatin PC2 across Model Phospholipid Membrane

Using electrochemical cell with two compartments separated by phospholipid membrane on polycarbonate support, transport of phytochelatin PC2 by means of cell penetrating peptide TP10 and in lesser extent with Mastoparan X was confirmed with voltammetry. Formation of the membrane and transport was followed by electrochemical impedance spectroscopy.
The Influence of Labile Complexes on Cadmium Transport across Artificial Phospholipid Membrane

The influence of labile cadmium complexes on transport across phospholipid membrane with A23817 ionophore – the increase of the amount of transported cadmium ions - has been found when only diffusion controlled transport was applied. Lability of Cd-MA complex was demonstrated with stripping chronopotentiometry. Used experimental setup can be utilized for specifying conditions for transport of metal complexes with different stability.
Characterization of Liposomes Used as Model System of Biological Membranes by Glassy Carbon Electrode

This contribution deals with study and characterization of liposomes. These vesicles may serve as model membranes used for study of transporting processes of heavy metal ions across the real phospholipid membranes (PLMs). 1,2-dioleoyl-sn-glycero-3-phosphocholine was used for the formation of large unilamellar vesicles (LUV). These vesicles were prepared by hydration method. Interactions of cadmium ions with LUV in solution have been characterized using voltammetry, where glassy carbon electrode was used as the working electrode. Size and presence of liposomes in sample was proved by dynamic light scattering and free cadmium ions in samples were determined by differential pulse voltammetry.
Characterization of Electrochemical Behavior of Lecithin-Cholesterol Mixture in Formation of Model Phospholipid Membranes

Electrochemical behavior of lecithin-cholesterol mixture in the form of phospholipid membranes (PLMs) was studied in this contribution. Lecithin (phosphatidylcholine) and cholesterol were chosen for this research, because they are essential for living cells. Electrochemical behavior of this system was studied using an electrochemical cell developed in our laboratory and with applying electrochemical impedance spectroscopy (EIS). Different ratios between lecithin and cholesterol placed on different polycarbonate substrates were tested. The influence of time and of temperature on formation of PLMs in presence of cholesterol was investigated.
Influence of LMWOAs Transport of Cadmium and Copper Ions Across Phospholipid Bilayer Using Calcium Ionophore A23187

The influence of oxalic, citric or malic acid on transport of cadmium and copper ions in dependence on pH has been studied. Artificial phospholipid bilayer containing wide-spectrum ionophore was prepared on polycarbonate support and its formation and stability was followed with electrochemical impedance spectroscopy. The species, transported across phospholipid bilayer were determined with voltammetry. The substantial influence of complexes formed with metal ions and LMWOAs on their transport has been confirmed.
