Reproductive isolation between diploid and tetraploid cytotype of Vicia cracca and possibilities of evolution of this aggregate
Vlčková, Zuzana ; Münzbergová, Zuzana (advisor) ; Mráz, Patrik (referee)
Master thesis investigates reproductive barriers in diploid-polyploid complex of Vicia cracca. Complex with basic chromosome number x=7 consists of diploid (2x=14), tetraploid (4x=28) and rare triploid (3x=21) cytotype. I studied prereproductive barriers between diploid and tetraploid cytotype: phenology of flowering, pollinators' behavior (preference of spieces of pollinators to cytotypes, sequence of visited cytotypes), variables, that could explain pollinators' behavior (amount of nectar as the main reward, size and amount of pollen grains as a potentional reward). To find out how strong the triploid block is I analyzed ploidy of seeds and seedlings from mixed-ploidy population. The habitat isolation showed up to be the strongest reproductive barrier. Pollinator's behavior meaningfully contributes to isolation, phenology of flowering contributes only minimally. Index expressing rate of prereproductive barriers is 0,956. Pollinator Bombus pascuorum visited on one locality preferably tetraploid plants and Andrena sp. preferred diploid plants. Even though tetraploid plants produce more nectar, no other analysis showed pollinators' preference to tetraploid plants. I prepared one squash of diploid V. cracca using method of in situ hybridization. This method needs to be optimilized for the studied taxon.

Complex with basic chromosome number x=7 consists of diploid (2x=14), tetraploid (4x=28) and rare triploid (3x=21) cytotype. I studied prezygotic barriers between diploid and tetraploid cytotype: phenology of flowering, pollinators' behavior (preference of spieces of pollinators to cytotypes, sequence of visited cytotypes), variables, that could explain pollinators' behavior (amount of nectar as the main reward, size and amount of pollen grains as a potentional reward). Pollinator's behavior meaningfully contributes to isolation, phenology of flowering contributes only minimally. Index expressing rate of prereproductive barriers is 0,990. Pollinator Bombus pascuorum visited on one locality preferably tetraploid plants and Andrena sp. preferred diploid plants. Even though tetraploid plants produce more nectar, no other analysis showed pollinators' preference to tetraploid plants.

Use of In Situ Hybridization to study evolution in plants
Vlčková, Zuzana ; Münzbergová, Zuzana (advisor) ; Šurinová, Mária (referee)
The main focus of this thesis is in situ hybridization method and its impacts in modern cytology. The first chapter of the thesis describes the method itself and its types, defines and enumerates fundamental terms of the method, describes the steps of preparation and actual experiments in detail and explains the process of evaluation and interpretation of results. Following chapter describes several cases studies on plants performed using ISH. It describes use of ISH to recognize introgression lines among hybrids and to determine their fertility, to study polyploids and evolution of the nonaploids and to study repetitive sequences in telomeres of lily plants. In the next chapter the thesis describes Vicia cracca agg. from morphological but mainly systematical point of view. The thesis is concluded by suggestion of possible utilization of genomic in situ hybridisation in order to reveal the pregenitors of tetraploid V. cracca and the evolution within the aggregate.

Nowadays, video games are a big part of mobile phone software industry. Special place holds multiplayer video games, which can be divided to short-range and long-range. This work aims to short-range wireless network technologies as Bluetooth, WiFi and Infrared. The main goal of the work is to create a real-time multiplayer variation of Bomberman game based on universal network interface in that way, so it will by totally independent on particular wireless network technology. Universal communication interface, will be implemented by a separate module for each network technology. Therefore a deep understanding of short-range wireless communication is required, which the focus of this work is also targeted on.
Recording legal relations and other data to lands in the real estate register
Vlčková, Zuzana ; Drobník, Jaroslav (advisor) ; Franková, Martina (referee)
THE ABSTRACT This thesis deals with the question of filing legal relations and other data to real estates with recording these legal relations and other data in the real estate register, included some problems which are in the real estate register incident and incurred while keeping the real estate register's state of filing in accordance with reality. To better realization of a generic context, especially for comprehension of the real estate register principles and recent state of filed data in the real estate register, there is also the historical evolution of registering the real estate in this thesis mentioned. It is difficult to understand some specific problems without knowledge of history because these problems result from former methods of registering the real estate and legal relations to them. Great attention is paid to keeping the state of filing in the real estate register in accordance with reality that is why the illustrations of some particular cases and examples are shown. This was led by effort to connect theoretical principles with their practical use, the major model situations were assumed from the experience of Klatovy Real Estate Register's employees. In the conclusion, an evaluation of the current state of filing in the real estate register is provided and there are some prospects...
Utilization of micropreparative isoelectric focusing in the analysis of biological samples
Vlčková, Zuzana ; Flodrová, Dana (referee) ; Šalplachta, Jiří (advisor)
The aim of this Master Thesis is the optimization and verification of conditions of separation of proteins from biological materials. For the separation a developed technique called micropreparative isoelectric focusing is being used. The theoretical part describes analytical methods of biological materials with a focus on proteins which were used for the optimization of the developing system in general. This part of the Master Thesis also presents this method itself. Furthermore, basic characteristics of other techniques used to control the efficiency of the micropreparative isoelectric focusing are introduced. The experimental part depicts the individual steps of optimization of the micropreparative isoelectric focusing. The ideal separation procedure was firstly found out on the individual standard proteins, secondly on their mixture and lastly the optimised procedure was applied to a real biological sample. The efficiency of the micropreparative isoelectric focusing was verified by SDS-PAGE and MALDI-TOF mass spectrometry.
Virtual playground
Svoboda, Tomáš ; Pelikán, Josef (referee) ; Vlčková, Zuzana (advisor)
Present document describes algorithms used in program Virtual playground. This program was designed to simulate movement and interaction of perfectly rigid bodies in real environment with gravity. It had to be optimised to be able to simulate tens of bodies on contemporary PCs in real-time. It was in fact also to the prejudice of physical precision but I think that the result is physically plausible. There is also vehicle simulation added to the program. I will try to introduce difficulties I had during programming and I will do a comparsion with other algorithms that are used to solve similar kind of problems. In the end, I will describe program structure and provide user guide for the program.
Configuration and routing mechanisms behaviour in ad-hoc
Vlčková, Zuzana
Technologies of local and personal wireless networks allows building ad-hoc networks which don't require prepared infrastructure of optical or metalic wiring. Besides mechanisms of channel sharing and mechanisms of routing, also mechanisms of topology control are very significant, in these networks. The goal of this diploma project is to evaluate behaviour of chosen mechanisms for topology configuration of wireless ad-hoc networks, optimizing suitable criterion - required energy or interference. Real node distribution of nodes in wireless networks doesn't agree with uniformly or Poisson distribution, published solutions are related to. Interesting would be to evaluate methods minimizing interference for more realistic node distribution (groups of nodes) and also in case of using frequency multiplexing.
