Molecular mechanisms of Wnt signalling in mammalian cells

Wnt signalling represents an important mechanism participating in control of cellular and developmental processes, including establishment of cell polarity, cell fate specification, stem cell self-renewal, tissue patterning and organogenesis, homeostasis maintenance and regeneration. Misregulation of the Wnt signalling during embryogenesis leads to developmental defects while aberrant activation later in development is associated with degenerative diseases and a number of cancers. The presented PhD thesis is based on four original publications that deal with the post-translational modifications of Wnt ligands and molecular mechanisms contributing to the regulation of a transcriptional profile of the so-called canonical Wnt pathway. Wnt signalling pathway is used repetitively both in time and different cellular contexts throughout development of multicellular organisms. Inevitably, in each single situation -catenin/TCF complexes, the downstream effectors, induce only subsets of all potential target genes. How this differential tissue- and stage-specific control over various subsets of target genes is achieved with such a limited number of nuclear effectors is not fully understood. Reception of Bedřich Smetana's Operas in Jan Malát's Arrangements

The thesis called Reception of Bedřich Smetana's operas in Jan Malát's arrangements deals with the typical phenomenon of 19th century musical life, e.g. music mediation in the form of arrangements. The thesis concentrates on a promoter Jan Malát and his arrangements of Bedřich Smetana's music. Jan Malát's arrengements played a substantial part in spreading Smetana's work. The thesis researches into Malát's nature of musical arrangements in relation to the original compositions of Smetana's operas. It also concentrates on changes in the society leading towards increase of demand for accessible music as far as the difficulty of interpretation is concerned. Based on the research of Bedřich Smetana's opera works reception, the thesis endeavours to describe reasons for publishing particular arrangements.

Crimes of Communism: "Work camps at urnium mines in Jachymov and Pribram in 50 Between the 20th century"

Crimes of Communism: "Forced labor camps at uranium mines in Jachymov and Pribram in Fifties of the 20th century " In this thesis, I tried to submit, if possible, factually true and correct image of a politically and legally difficult period in our postwar history. Communist revolution in February 1948 started the socialist reconstruction of our society with all the attendant phenomena of rising totalitarian power. Persecution of political opponents and their re-education and forced labor work in uranium mines in Jachymov and Pribram is a really sad reality of our recent history of the fifties of the last century. In retrospect, absurd crimes these prisoners and incredibly inhumane prison conditions and rules of inconvenient people in forced labor camps only show monstrosity of Communist ideology and are proof of loathing practices then representatives of the ruling party. Overview of forced labor camps, the operation of the communist judiciary, the number of incarcerated people, and unfortunately, the numbers who stay in the camps and work in uranium mines have not survived are so by drawing on what the historical stage brought our ancestors and what must remain unforgotten.

Prevention of complications in thyroid surgery

This thesis presents a retrospective analysis evaluating incidence of early postoperative complications of total thyreoidectomy and hemithyreoidectomy using conventional surgical techniques and harmonic scalpel. The first part describes the history of surgery of thyroid gland, its topography and indications. It also describes surgical procedures on the gland, along with possible postoperative complications and possibilities of their prevention. It summarizes development of new methods and instruments in thyroid surgery in recent years. The second part compares the aforementioned surgical methods to assess contribution of harmonic scalpel as a potential tool for prevention of postoperative complications.

Comparison of Selected Methods of Valuation of Garage-type Real Estate in the Locality of České Budějovice

This thesis compares the selected type of property valuation methods in the garage area of the Czech Budejovice. Selected garages were awarded cost method, comparative method and comparison method from the database. At the beginning of the thesis describes the basic concepts of real estate valuation. The following describes the operators occurring in the real estate market, a general description of the garage. To conclude the theoretical part describes selected methods of evaluation of existing garage, according to the rules and regulations according to previous regulations. For the purpose of this thesis was awarded with 10 garages, of which 8 are terraced and 2 detached. Learning rates are compared and statistically analyzed.

How non-price factors affect the attendance of the Museum of Eastern Bohemia in Hradec Králové?

This thesis analyses non-price factors influencing the attendance of the Museum of Eastern Bohemia in Hradec Králové. On the basis of daily data, from 05/2011 to 05/2013, using average daily temperature, total daily precipitation, marketing, work and school holidays and individual months, three models were construed -- a model of total attendance, attendance of adults and attendance of children and students. Research has proven expected negative influence of temperature through all three models. Furthermore, there was a strong positive influence of school and work holidays. The results have also shown almost 50% increase of student attendance during holidays. Similarly, attendance of adults increases even more during non-working days. Marketing was studied in different ways but its statistically relevant influence has been proved only in the groups of children and students. Their attendance increases by quarter during campaigns.
