Completion of the BUT - Faculty of Fine Arts

The topic of my master´s thesis was to create an architectural study of the Fakulty of Fine Arts in Brno - city part Veveří. The site lies near the Faculty of Law of Masaryk University and it´s bordered by streets Šumavská, Veveří, Bulínova and Akademická. The project ties together the architectural study from subject TG02 witch solved the second part of the programme "Academic Square" and it was a construction of the fakulty of Forensic Engineering and the background for both faculties. So the solution is a compex architectural study which sorts the urban and also the architectural aspects of both faculties and their background. The main idea of the urban concept is to také up the surrounding bulit-up-area which is typical for this part of Brno. The philosophy of the architectural solution comes from the concept of the traditional British universities. So the solution are two objects of the faculties with plane shape od the letter U which clutches the volume of the common background between each other.
Detached house with a car repair shop

The bachelor thesis deals with design house with a car repair shop and elaboration of design documentation of the building. It is a slab-brick building, which is divided into a two- story living area and single storey part of the car repair shop. The building will be covered with flat and gablet roof. The main text contains the message type A, B, D. The content is guided by Annex 6 of Decree No. 62/2013. Attachments are inserted into the preparatory work, the drawings and calculations. Annex consists of 7 components.

The bachelor's thesis Plant motif in painting at the end of the 19th and the first half of the 20th century consists of two parts, theoretical and practical. The theoretical part deals with the motif of plants in paiting at the end of the 19th and the first half of the 20th century. It introduces flowers as plants in the layers of meaning that are part of the artwork. As an example is presented the work of P. A. Renoir. The practical creative part presents eight wash drawings with plant motifs, such as: gladiolus, lily, hydrangea, rose, sunflower and zinnia. The presentation includes a series of sketch material.
Development of Educational System in Pelhřimov Region with Concentration on a Small School with Mixed-age Classes in Častrov village

1 ABSTRAKT The aim of the thesis was to eleborate the development process of the Častrov comprehensive primary school in the district Pelhřimov based on the school chronicles and refer to the contribution of the acquired knowledge for the schoolwork. The theoretical part is devoted to the study of commemorative books and also in the wider perspective of the whole P elhřimovsko region. The practical part describes a project carried out with pupils of this school. The purpose was to acquaint the pupils with the history of Častrov school through activation methods and have their impact on the school and the place where they live.
Proteomic analysis of cellular proliferation and differenziation: Model of neural stem cells and cancer cells

CoNcLUsIoNs In protein profiling of neural stem cells using 2-dimensional gel electrophoresis and mass spectrometry, constitutively expressed proteins in 66 protein spots were identified. Most of the individual protein species were related to RNA and protein metabolism, processing and turnover, including some chaperones and stress response proteins. Proteins involved in cellular organization (e.g. cýoskeletal proteins and annexins), metabolic proteins (mostly enrymes),cellular energetics,cell defenseand signallingfollowed in lower numbers. Proteins in 16 spots significantly regulated during neural differentiation were identified. Induction of levels of o-B crystallin, hnRNP Al and hnRNP AZIBI during differentiation and protein localization within neural cells were studied by westernblottingand immunocýochemistry. Using antibody microarrays, in neural stem cells an increase in GRK2 level and phosphorylationsof signalling molecules(CDKI|Z, PKC mu, PKCy, Erk5 and o-B crystallin) involved mostly in cellular proliferation were detected.On the contrary, in differentiatedneural cells levels of protein-phosphatase4, heme-oxygenase2, MEK3, RafB, pro-caspase 1 and phosphorylation of 40 kDa proline-rich Akt substratewere induced. Proteomics as a tool for understanding molecular mechanisms of human diseases

Proteomics is a set of analytical methods which enable qualitative and quantitative characterization of the proteome. Expression proteomics quantitatively compares proteomes of cells, tissues, body fluids or other biological materials to find differencies in protein expression and, based on these differencies, to describe the biological processes occuring in investigated organisms. An initial material for expression proteomic studies are complex mixtures containing thousands of proteins, which are analyzed using separation (electrophoretic and chromatographic) methods, and identified, possibly quantified using mass spectrometry. The aim of this Thesis is to demonstrate the application of the tools of expression proteomics in solving diverse challenges in biomedicine. We employed various proteomic approaches and tools for studying molecular mechanisms of human diseases using pacient biological samples, or a model organism and a cell culture. Proteomic approaches in cancer biology

Proteomics as a modern comprehensive approach to the analysis of proteomes was applied in three projects aimed at diagnosis and therapy of cancer. The aim of the first the project was to find a new diagnostic biomarker for ovarian cancer. Two different comparative proteomic approaches were used for comparative analysis of sera from patients diagnosed with ovarian cancer and from healthy age-matched women. We identified -1-antitrypsin with increased concentration in patien sera, and apolipoprotein A4 and retinol-binding protein 4 (RBP4) with significantly decreased concentration in patients. The significantly decerased concentration of RBP4 in patients is a new observation. We propose that RBP4 is either decreased in ovarian cancer patients as a result of its reduced production by ovary or it may reflect less specific systemic changes, for instance early onset of cancer cachexia. The second project was focused on gaining insight into the molecular mechanism of cytarabine resistance in mantle cell lymphoma (MCL). Proteomic and transcriptomic analyses of cytarabine-resistant cells revealed marked downregulation of deoxycytidine kinase (DCK) - a protein essential to intracellular activation of purine and pyrimidine nucleosides and their analogues including cytarabine.
