Analysis of co-transcriptional spliceosome assembly on RPL22B

In eukaryotic organisms, the process of splicing is used to remove non-coding sequences - introns - from transcripts to form mRNA, and together with transcription, it also allows to regulate the amount of the resulting RNA that will be used further in translation. The protein of interest in our study is Rpl22, which binds the intron of its own transcript and its paralog and can inhibit their splicing. This thesis examines the mechanism by which the inhibition occurs and provides new information about the regulation of RPL22 transcript expression via its intron. Using co-transcriptional spliceosome assembly analysis on the RPL22B transcript, we were able to observe that the recognition of splice sites by the U1 snRNP, Msl5 and Mud2, is not affected by the inhibition. Thus, the inhibition of RPL22B splicing will occur later during spliceosome assembly or during spliceosome activation, before the first step of splicing takes place.
How social networks affect celiacs'diets

This paper examines the influence of social networks on the diet of celiacs. The aim was to investigate how social networks, specifically Facebook, Instagram, YouTube, Twitter and Pinterest, can influence the preparation, purchase and consumption of gluten-free products. The chosen research method was quantitative analysis. Results were obtained from Czech and a few Slovak celiacs and people with gluten allergy with help of a questionnaire survey followed by descriptive analysis. The questionnaire itself was disseminated on the social networks Instagram, Facebook and WhatsApp. The final sample included 264 from the original 279 respondents. The results show that the use of social networking sites is popular among patients with celiac disease and gluten-free diets and that users benefit from it. It was also revealed that the most useful social networks are Instagram amd Facebook. On the other hand, YouTube, Twitter and Pinterest fell completely in the evaluaion. The main reason for this is the small amount of beneficial information. When it comes to eating, celiacs are mainly looking for new recipes, products and places that offer gluten-free meals, according to the results. Populace hnědásků (Melitaeini) na nelesních stanovištích NP Podyjí refaunovaných pastvou koní

This thesis evaluates the impact of refaunation by the Exmoor pony on populations of five Melitaea butterfly species - Melitaea athalia, M. cinxia, M. didyma, M. britomartis and M. aurelia inhabiting xeric grasslands of the Podyjí National Park - Havranické Vřesoviště and Mašovická Střelnice. Both grassland sites are grazed since 2018. I compared adult butterflies' demography structure, mobility, spatial distribution, and habitat requirements with an earlier study carried out before the impact of horses (Vodičková et al., 2019). I detected multiple changes, some of them attributable to the horses (e.g., butterfly spatial distribution) and some not (e.g., population sizes), but also a remarkable stability, e.g., in dispersal parameters.
Creating of local network of community midwives (Action research)

This thesis is based on action research which sits at the interface of midwifery and the management of health and social organizations. It maps the development of a local network of community midwives in Brno between 2021 and 2022. In the beginning, a Brno community midwife had a dream to work in a birthing house (midwifery unit). However, as this is currently impossible in the Czech Republic, she looked for different ways to work with her colleagues to provide accessible and quality care to women who demanded it. Thanks to a grant from the Brno-Centre district, five facilitated meetings of midwives were held. These meetings explored the motives and needs of midwives, the obstacles they face and their experiences in their practice. In this way, a service was being gradually designed that could provide women with individualised comprehensive support and care during pregnancy, birth and the postpartum period, provided by midwives. Today, the original dream has taken concrete form - Nest. It is a (for now informal) group of midwives who meet regularly, have a defined common vision, shared values and principles of care, collect statistics from births, have regular supervision and gradually implement a system of substitution. Title: The relation of laterality and selection of rotation direction in figure skating at children in preschool and younger school age Objectives: The aim of the thesis is to construct a test battery on the ice surface, to assess the laterality, i. e. the preferred lower limb and the preferred side of rotation and turning, for beginning figure skaters in preschool and younger school age. Subsequently, individual items of this test battery are examined with items from the standardized test battery for the child population aged 8-10 years, according to Musálek (2013). The relationships of individual test items are determined in order to determine whether the individual test items on the ice surface can be used to represent a relatively reliable tool for determining the rotation direction of a beginning figure skater. Methods: We use testing by standardized battery of tests according to Musálek (2013) for the diagnosis of motor manifestations of laterality for children aged 8-10. Vaccination as a communication challenge: an analysis of the arguments and factors influencing its (non-)acceptance

This diploma thesis deals with the negative phenomenon of refusing vaccination,the declining vaccination coverage, and the return of infectiousdiseases which are preventable by vaccines. This is an international phenomenon which was identified by the World Health Organization in 2019 as one of the top ten threats to public health. The topic of rejection and hesitation in vaccination is a complex issue which is influenced by many factors. These include,for example, the media environment, geographical and financial barriers, historical influences, personal experience, health literacy, risk perception and many more. Failure to adopt immunization programs is a collective, not individual, public health problem. It is therefore necessary to map these factors and to analyze the arguments against vaccination which prevent its adoption. The thesis contains a brief mapping of the history of refusal of vaccination, elaboration of a comprehensive set of factors influencing the acceptance of vaccination, analysis of selected arguments against vaccination, and finally, a summary of knowledge for creating strategies for adopting immunization programs.
Development of kinetic habits and skills within Prague schools children

The aim of my thesis was to create motor tests and to chose individuals, who did these tests, according to different criteria. My thesis consists of a theoretical part and practical part. In the theoretical part, I have summarised the issues of physical activity and other related topics. In the practical part I have used a testing method. After carrying out the tests with selected individuals I analysed the results. I compared them with specialized literature (see Měkota) and evaluated them, whether they correspond or not with the correct motor development of children in various motor skills ( strength, speed, endurance, dexterity and agility). Crisis of informal carers in the care of seniors at home

This thesis deals with informal (family) carers who provide for the care of an elderly person in their home environment. It focuses on selected moments of crises (motivation, accepting support, financing, impacts) based on the analysis of documents and the author's two-year working experience in this field. The objective of this thesis is to find out how the carers cope with such crises and what strategies they choose in their provision of care. The theoretical part of the thesis defines basic terms related to old age and their values, informal carers and related areas of care, including pre-defined crises. The research part is divided into methodology, in which part the author describes her methods of obtaining data through semi-structured interviews; and into the empiric part, which is based on the statements of the carers interviewed. Development of Motor Skills of Primary School Children Assessed by the Test of the School Readines

The purpose of my research was to determine the level of motor skills and school readiness in preschool and primary school children. The thesis consists of theoretical and practical part. Theoretical part deals with development of motor skills, school maturity and characteristics of preschool and primary school period. Practical part includes the results of testing fine motor skills and rough motor ability using the assesment of the test of the school readiness. The results for children of different age were mutually compared and then compared with professional literature. Key words: Motor skills, motor development, school readiness
