Treatment of spinal cord injury by transplantation different types of stem cells

Spinal cord injury (SCI) is complicated injury with serious socioeconomic consequences for the patient and his whole family. Big difficulty cause also extremely high living expenses for the patient with this type of injury. That's why there is a need for therapeutic methods which would help patients after SCI to recover the lost functions and be able at least partially to return to their normal life. Different therapeutic methods are being used for SCI treatment. In this study we used four various types of stem cells: human bone marrow stem cells (hBM-MSCs), human umbilical cord mesenchymal stem cells (hUC-MSCs), neural precursors derived from induced pluripotent stem cells (iPS-NPs) and neural stem cell line derived from human fetal spinal cord tissue (SPC-01). These cells have been transplanted intrathecally or intraspinally 7 days after induction of the experimental model of SCI in the rat. We studied expressions of genes related to neurogenesis, growth factors and inflammation 10 and 28 days after SCI. Our analysis showed significant changes in gene expression 10 days after SCI. Moreover, cells experience distinct microenvironmental features within the extracellular matrix in tissues, that 3D cell culture might replicate. In fact, majority of studies indicate that physical cues generated from extracellular microenvironment drive the cellular behavior and functions. In this dissertation thesis, we attempt to provide the mechanistic explanation behind the changes in cellular metabolism and molecular signaling pathways induced by physical cues of 3D cell culture. OUTSIDE LIVING ROOM - REVITALIZATION OF URBAN COURTYARD

Main aim of the thesis is to create an outdoor living space inside of tradition housing blocks. Principles of placemaking were used during a design process. One of the main ideas is to return a water to the urban courtyard according to its history.

The topic of my thesis is the student center at Poříčí - on the corner Táborského nábřeží and street Vídeňská. It proposes seven-storey building containing student housing, student`s canteen, sports facilities and a calm inner courtyard. The building is addressed clearly and simply. It closes to the busy Street Vídeňská and it opens towards the River.

Completion of the area Údolní 53 for the Faculty of Architecture and Faculty of Fine Arts

It is necessary to ask the question of the relationship and the corresponding forms of buildings of the faculty of architecture and faculty of fine arts with the public. Due to the efforts of the faculties to open up to the public, I find it undesirable to place new objects in the courtyard and hide behind the walls of historic buildings. Much more meaningful is the use of existing buildings at the boundary of the plot. It is important to protect our history, but the protection of existing buildings should not get to the point where it will defend the presence, be on the same level, and thus ensure its quality and together to shape the future. The newly allocated creative use of the original barracks has greater demands on space, whether capacity or its generosity. Therefore, it is necessary to customize the objects for the current function. The superstructure will help keep faculties in one building. An advantage is also the connection to existing networks or vertical communications. The proposed minimalist intervention is in contrast to the historic building, making it stand out, while the verticalism of the superstructure raises the object of the original barracks to the desired institution of the Faculty of Architecture and the Faculty of Fine Arts.
Socio-cultural centre Dolní Kounice

My diploma thesis is based on the module Res Publica - Public works focusing on community buildings. Dozens of towns were surveyed, whether they are interested in reconstruction of their community or sociocultural centre, and eventually a small town Dolní Kounice was chosen especially because of the beauty of the surrounding countryside as well as the exciting native urbanism and architecture. This thesis deals with transformation of the cultural house from the 50s of the 20th century to a new cultural centre. The extension and inconvenient terrace were removed and the hall is supported by a new mass of the vestibule at the interface between interior and exterior, on the site of the former patio. Next, a new front of the building was introduced which increased the entrance of the house and created a space for a study room, small gym, meeting room and common rooms. The part of the house used for housing units is designed only with aesthetic details on the facade. The Culture Centre is divided into a set of materials in order to reduce the scale of the whole building. The entrance part mass, in contrast, is enhanced by its division and the chosen materials. In this way the originally embarrassing object of the Cultural Centre has been changed into a new centre with a clear architectural expression and the interior of the house is enriched by view of the beauty of the city.
Residential Complex in Brno - Komín

This design is based on the site's strong potential in regards to its attractive location near Brno city centre and at the same time its proximity to nature. The solution of volumes results from an effort to create a system of public through private spaces while maintaining the continuity of the whole. This residential complex can offer apartments of all sizes, from flatlets to five bedroom flats and even houses. It also provides its neighborhood with new possibilities to shop and to spend time outdoors.
Living by the river

The city of Brno is the capital of Moravia in the Czech Republic which was shaped by many historical events, its predominant textile industry was amongst the most important aspects of these criteria, since its main rivers cut through the city (Svratka, Svitava), this industry and its trade, was a main factor in shaping the city as a whole, alongside political conflicts, change of regimes. During the 1960s, after the planned style economy was settled completely, it resulted in big setbacks for the Czechoslovak economy, as the textile industry started to crumble. The final blow to this industry happened after the velvet revolution as privatization took place and most remaining factories were dismantled. This narrative had a big role in shaping how Brno looks today as the northern part of the city is more urbanized than the southern part which is more industrialized by nature, and unlike Prague, these two main rivers are not widely crucial to the public life. This also developed along with other factors a major problem with accommodation quality, price as official data reports increasing dissatisfaction from inhabitants with housing.
CERTIFIED METHODOLOGY, Methodology for low-volume sterilization hatching eggs

The methodology is the result of the solution of the research project No. TH04030206 entitled: Develoment of methods of physical sterilization of hatching eggs. The project is focused on a hatching egg sterilization based on physical methods - namely low temperature plasma (LTP). Compared to the existing methods, LTP should result in an increased efficiency of poultry farming and eliminated health and ecological risks. The higher added value and efficiency further contribute to higher competitiveness of the poultry industry and food self-sufficiency of the Czech Republic.
