Evolution of karyotype, sex chromosomes and gene clusters in selected taxa of araneomorph spiders

Spiders (Araneae) are a diverse group with various sex chromosome systems. The most common is X1X20 sex determination, X1X2 denotes two non-homologous X chromosomes and 0 the absence of Y. The presence of Y chromosome is rare among spiders and has been described mainly in Synspermiata, X1X2Y is probably ancestral constitution for this clade. The family Sicariidae was chosen to elucidate the evolution of sex chromosomes in Synspermiata. Karyotypes of 10 species whose 2n ranged from 19 to 23 were investigated in this study. The most common finding was X1X2Y in all studied Loxosceles species. The derived XY system was discovered in Sicarius. Furthermore, the distributional pattern of molecular cytogenetic markers (genes for 18S rRNA, 5S rRNA, histone H3 and U2 snRNA) was analysed using fluorescence in situ hybridization (FISH). These markers were found exclusively on autosomes, on one or two pairs. FISH with whole-genome probes of both sexes was performed to reveal distribution of Y specific sequences. Rapidly mutating Y-STRs analysis as a tool for forensic investigations, genetic genealogical applications and molecular anthropology

Compared to standard Y-STR markers commonly used in forensics and bioarchaeology, rapidly mutating Y-STR markers (RM Y-STR) have a significantly higher haplotype diversity and lineage resolution abilities in world populations. They form a novelty in these fields. This paper will attempt to clarify their origin, function, and efficacy in forensic genetics, genealogy, and molecular anthropology by looking at the Y chromosome and its characteristics that allow its use in the mapping of populations, individual male lineages, and identifying male perpetrators of sexual violence. By looking at other polymorphisms of the Y chromosome, especially Y-SNP markers, we can see in which studies each type is used most effectively. In the end, it will be clarified whether the use of RM Y-STR markers is beneficial to each field, whether they make standard Y-STR markers redundant, or whether they are themselves of no substantial use in comparison.
Y chromosome in the mouse hybrid zone

The contact zone between subspecies of house mouse Mus musculus musculus and Mus musculus domesticus is one of the most intensively studied hybrid zones. It is also due to extensive introgression of the Y chromosome of M. m. musculus subspecies to the genetic background of M. m. domesticus. One theory of the origin of the introgression explains it by intragenomic conflict between the sexes. With a set of variable microsatellite markers on the Y chromosome, I have examined the validity of this theory by simple approaches revealing the history of the introgression area. It turned out that overly big variability of our markers makes the revelation of this theory impossible. Our markers have been found suitable for use in the analysis of population structure of house mouse. Thanks to them, we can identify migrants between localities and estimate the level of closeness of the population structure in relation to migrants from the neighborhood. Populations in our analysis proved to be relatively closed and resistant to the influx of migrants. Despite the conclusions of previous research where the dispersion of males ran up to one kilometer, I have discovered a relatively large number of migrations to a distance of thirty kilometers. SNP polymorphisms of Y chromosome in the population of african fulani people

Markers on the non-recombining region of chromosome Y is a useful tool for study of diversity between populations. SNPs are the most commom polymorphisms in human genome. Mutation rate of SNPs is very low and so they may be used as genetic markers in evolutionary and population studies. We have analyzed 205 unrelated men from 11 Sub-Saharan Fulani's subpopulations. Fulani are an ethnic group of people spread over many countries, mainly in West Africa. Our samples are from Tindangou area, Banfora area (Burkina Faso), Bongor area, Linia area (Chad), Diafarabé area (Mali), Tcheboua area (Cameroon), Banfora area, Diffa area, Zinder area, Ader area and Abalak area (Niger). Using kit Signet Y-SNP Identification Systems and Luminex instrument with LabMAP Luminex Technology we detected particular Y chromosome's SNPs. LabMAP Luminex Technology is universal array platform, which as a probe using fluorescent polystyrene microspheres. We have observed 12 different haplogroups. Haplogroup E, which is typical African haplogroups, is determined with derivated allele in polymorfism M96. We have detected haplogroup E in maximum of 89,3% in the Fulani's subpopulations. The impact of geography and subsistence on distribution of NRY haplogroups in Europe and Africa

Y chromosome is due to its special characteristics the ideal tool of archaeogenetic studies. Its diversity is influenced by several factors and I analysed two of them (geographical location and subsistence). I generated SNPs and STRs data from several loci of samples from Slovakia (156 samples, 5 regions) and sub-Saharan Africa, where I analysed samples of sedentary farmers (481 samples, 18 regions) and nomadic pastorals (405 samples, 16 regions). Slovakia is situated at the meeting point of two migration ways. First of them was spread from the east to the west and is associated with enlargement of haplogroup R1a in Europe. The second came from the Iberian Peninsula eastward and is associated with enlargement of haplogroup R1b. Results of MDS graphs replicate the geographical map of Europe. Slovakia is situated in the middle of Russian, Balkanian and Iberian samples. Correlation between genetics and geographic distances is indicated by hierarchical AMOVA analysis and Mantel tests. Populations in sub-Saharan Africa differ from each other by the subsistence pattern. Different life style influence the diversity of the Y chromosome. Population structure, migration and dynamics in Africa and Arabia

In addition to the interaction of evolutionary forces, the population history of the African Sahel and Arabia has been influenced by the spread of Neolithic cultural innovations. The reflection of these processes today is a very complex structured diversity of the current populations, which is presented here through the analysis of several genetic markers. The aim is to provide a comprehensive view of the history of demographic processes in the Sahel and Arabia, by combining genetic, linguistic, subsistence and geographical data obtained from local populations. A study of a large dataset of mtDNA sequences showed that Arabia was a major crossroads in gene flow, and although it was colonized by anatomically modern humans from East Africa, today's differentiation from Africa is greater than the differentiation between local populations in these regions. Even the Sahel was an important biocorridor in the past. Today, we encounter populations of various subsistence strategies (nomadic pastoralists and settled farmers), between which gene flow has been severely restricted. A comparison of uniparently inherited loci in both groups points to different migratory activity in the eastern and western parts of the Sahel.
