Analysis and characterization of antimicrobial peptides by capillary electrophoresis
Tůmová, Tereza ; Monincová, Lenka ; Čeřovský, Václav ; Kašička, Václav
Capillary electrophoresis has been employed for quantitative and qualitative analysis, separation and characterization of antimicrobial peptides – halictines. In addition to purity degree of synthetic peptides, also their effective and ionic mobilities and charges within a wide pH range were determined.
Rychlé a citlivé stanovení metforminu v lidské moči a séru pomocí kapilární elektroforézy s bezkontaktní vodivostní detekcí
Determination of chlorophenols in environmental samples using electromembrane extraction and capillary electrophoresis
Šlampová, Andrea ; Kubáň, Pavel ; Boček, Petr
Combination of electromembrane extraction (EME) with capillary electrophoresis (CE) was used for determination of trace level chlorophenols (CPs) in environmental water samples. The analytes were transported across supported liquid membrane (SLM), composed of 1-ethyl-2-nitrobenzene (ENB), by the application of electrical field. A driving force of 150 V was applied to extract the analytes from neutral sample (donor solution) into strongly alkaline acceptor solutions. The acceptor solutions were subsequently analyzed by CE with UV-Vis detector.
Low-cost 3D-printed fluorescence detector for capillary electrophoresis
Přikryl, Jan ; Foret, František
We have verified the possibility of Fused Deposition Modeling (type of additive manufacturing) for lab-made construction of inexpensive fluorescence detector for capillary electrophoresis using commercially available optical components. Sensitivity of fabricated and assembled detection system was tested by fluorescein solutions. Limits of detection were determined for several tested arrangements.
Capillary electrophoretic chiral analysis and estimation of helix inversion barrier of helquats using randomly sulfated cyclodextrins as chiral selectors
Kašička, Václav ; Koval, Dušan ; Sázelová, Petra ; Severa, Lukáš ; Adriaenssens, Louis ; Vávra, Jan ; Teplý, Filip
Capillary electrophoresis in acidic sodium phosphate background electrolyte, pH 2.4, with randomly sulfated alpha-, beta- and gamma-cyclodextrins as chiral selectors was employed for fast, high-resolution separation of enantiomers of a series of 24 helquats, a new class of N-heteroaromatic dicationic helicene-like species. The developed CE methodology will be utilized in the further investigation of helquats, e.g., in evaluation of their preparative enantioresolution procedures, in monitoring of their racemization and determination of interconversion barrier of helquat enantiomers.
A simple sample pretreatment device with supported liquid membrane for direct injection of untreated body fluids and in-line coupling to a commercial capillary electrophoresis instrument
Pantůčková, Pavla ; Kubáň, Pavel ; Boček, Petr
A simple sample pretreatment device employing extractions across supported liquid membranes (SLMs) was designed for in-line coupling to a commercial CE instrument. The device can be easily assembled/disassembled and SLMs can be replaced after each extraction thus minimizing sample carry-over, avoiding tedious SLM regeneration and reducing total pretreatment time and costs.
Capillary electrophoresis with contactless conductivity detection of cationic-derivatized oligosaccharides
Partyka, Jan ; Foret, František
The oligosaccharides were derivatized by quaternary ammonium label and separated by CE with contactless conductivity detection.
Elektroforetická separace oligosacharidů s vodivostní detekcí
Partyka, Jan ; Foret, František
Zlepšení separačních vlastností oligosacharidů pomocí derivatizace kvartérní amoniovou značkou.
Analysis of antimicrobial peptides by capillary electrophoresis
Ehala, Sille ; Niederhafner, Petr ; Čeřovský, Václav ; Řezanka, P. ; Sýkora, D. ; Král, V. ; Kašička, Václav
Capillary electrophoresis was used for the purity degree determination of new antimicrobial peptides: octadecapeptide melectin and tetradecapeptide mastoparan PDD-B, and their synthetic analogues. Besides, four semiempirical models correlating electrophoretic mobilities of these peptides with their charge and size (relative molecular mass) were applied to predict their probable structures in solution. Additionally, capillaries coated with gold nanoparticles were employed for the separation of mixtures of antimicrobial peptides.
Recent applications of capillary electrophoresis to analysis and physicochemical characterization of peptides
Kašička, Václav ; Koval, Dušan ; Ehala, Sille ; Sázelová, Petra ; Šolínová, Veronika ; Hlaváček, Jan
Recent developments in the methodology of capillary electrophoresis are presented with respect to their application to analysis, separation and physicochemical characterization of biologically active peptides. New strongly acidic/basic classical and isoelectric background electrolytes were applied to high-sensitive analysis and high-efficient separations of structurally related peptides, such as insect oostatic and antimicrobial peptides, phosphinic pseudopeptides and tryptic peptide fragments of proteins from native and genetically modified maize species.
