Allergens in the work enviroment

The bachelor thesis on allergens in work environment is divided into two parts. The theoretical part, which describes the issue of allergic diseases, allergens and occupational safety from the general point of view and the practical part, in which the issue of allergic diseases arising in direct connection with the working environment is displayed using the data obtained. Four basic research questions were asked: "Which allergic diseases were most common in the work environment?", "Which allergens were most common in the context of allergic diseases?", "Which type of occupation was most commonly associated with allergic diseases?" What is the trend in the development of occupational allergic diseases?" The other 2 questions were focused on the links between allergenic substances and the work sector in which allergic occupational diseases occurred and the links between allergenic substances and the specific diseases that occurred. The bachelor thesis has been handled as a secondary analyse of data, these data have been granted by a Regional Hygienic Station in České Budějovice, department hygiene in work place. All the data included in this bachelor thesis were concerned all recognized illness from work in the South Bohemian Region from 2010-2018.
Synanthropic arthropods as a producers of allergens in households

This work is about arthropods producing allergens in our homes. It sums together not just todays knowledge about allergic effects in human environment and possible diseases, but also about behavior, occurrence and life cycle of species themself and about possible elimination arthropods and allergens from our households. Most of the world population is nowadays affected by the allergies, allergens are all around us. In today's world when people spend most of the time in the interior enviroment whitch contains allergens and their sources, are also important allergic diseases caused by the arthropods, which we share our homes with. Almost in every room in our home are present some of the arthropods producing allergens. These allergens can affect the life of sensitive persons. Allergens are inside the body of the arthropods and in their body secretions. The highest volume of allergens coming from arthropods in our homes is contained in a dust and food. Allergic reaction is caused by consummation, inhalation or body contact. The cheapest and the most effective treatment of the allergic diseases is to avoid the allergens. The best what we can do is to regulate their volume in our households, because the complete elimination is practically impossible. Key words: Allergens, synanthropic arthropods, households
Alergens in the Teaching of Natural Science, Biology and Chemistry

The theme of the diploma thesis is allergens in the teaching of natural science, biology and chemistry. The subject and its aim is to find out the presence of allergens in selected classes of natural sciences (natural sciences, biology, chemistry) as well as the level of awareness of problems of allergies and allergens from teachers and pupils of elementary schools and grammar schools and to compare whether teachers of selected subjects take into account Creating climate climate for allergic pupils. The thesis consists of theoretical and practical part. The first part summarizes all theoretical knowledge, including the theme of allergies and allergens. It deals with the concept of allergy from the historical point of view, it characterizes the general importance of allergies and its connection with the immune system, it presents individual risk factors influencing the origin and development of allergic disease. A large part of the theoretical part describes allergic diseases including their manifestations, diagnosis and follow-up treatment, and also the individual types of allergens that are classified according to their effects on the organism. The content of aroma active compounds in sweets

This bachelor thesis deals with the assessment of aroma active compounds in non-chocolate confectionery. In the theoretical part the production technology of non-chocolate confectionery from raw materials to final product is described. Furthermore, aroma active substances, including potentially allergenic, are characterized, and the method for their determination by solid phase microextraction and gas chromatography with mass spectrometry is briefly described. The aim of the experimental part was identification of volatile substances and subsequent quantification of 24 potentially allergenic aroma substances by the HS-SPME-GC/MS method. Gummy candies, purchased in the regular market, were used as model samples. The differences in the number and type of volatile compounds were found between the samples. Overall 230 volatile compounds were indetified including 8 allergens. These were composed of 55 alcohols, 68 esters, 29 alkenes, 18 aldehydes, 25 ketones, 2 ethers, 3 epoxides, 9 carboxylic acids and 21 other compounds.
Comparison of cytokine production by leukocytes from newborns of healthy and allergic mothers

The increasing incidence of children suffering from allergic diseases could be caused by sensitization of immature immune system during the intrauterine development. Several important scientific papers have demonstrated the ability of cord blood cells to respond by elevated proliferation activity after stimulation by common allergens. Following these findings, present study follows the production of cytokines which play a role in the pro- and anti-allergenic tuning of the immune system. Umbilical cord blood cells were stimulated with polyclonal activators (phytohaemagglutinin) and common allergens (ovalbumin, timothy grass, birch, mite). Subsequently, cytokine production was monitored using selected methods that reflect different stages of cell activation - at the level of mRNA by quantitative real time PCR (qRT-PCR), by flow cytometry detection of the presence of intracellular cytokines in different cell subpopulations and by ELISA measurement of cytokines in CBMC culture supernatants. The results obtained point to a very weak ability of these common allergens (timothy grass, birch, mite, ovalbumin) to stimulate CBMC to produce cytokines observed by all of these methodological procedures. Diets for food allergies

I have chosen topic Diet with Food Allergy as a topic of my bachelor thesis. This illness is quite common these days. It affect infants mostly but allergy of cow´s milk protein of children under three years disappears itself mostly. There is increasing amount of causes when patient suffers more allergies. Number of people who have the most serious reaction the anaphylactic shock, icreeases. It can end with death. Doctors recommend to patients to strictly follow the elimination diet which is basic treatment of this illness. The most accurate diagnosis of illness (immunological or not) is another important factor for effective treatment. I have chosen three aims for this bachelor thesis. First to find out if patients are informed about their illness. Second to map eating habbits of people with food allergy. Third the evaluation of correctness of daily menus. I used approach of guality research. Research group consisted of six respondents with different food allergy. Those respondents wrote down their weeks menus. Menus were evaluated with program Nutriservis and it was assessed if it follows recommended dietary value. Suitable intake of energy and essential nutrients was calculated for every respondent. It was evaluated if respondents follows priciples of rational diet and if their diet contains sufficient amount of minerals and vitamins. Questionnaires were another key point of this research. Questionnaires were about illness know-how of patients and who has instructed them about it. It was found out the patients hasn´t got enough information about their illness and half of them has no idea which essential nutrients, minerals and vitamins could lack in their diet. It resulted from menus simultaneously that five of six respondents do not follow principles of rational diet and eat very monotonously. Only one respondent follows strictly elimination diet others violate diet exceptionally. Respondets would appreciate other research would focus on creation of special centres and training of doctors and nutritional therapist who would be focused on food allergies. Spreading the information about illness not only to patients but to public also is really important. Another important point is to increase amount of books about food allergies at the market.
Determination of alergenic and potential alergenic metals in cosmetics

The aim of the diploma thesis was to provide an overview of the prevalence of allergenic and potentially allergenic metals in eye shadows. The diploma thesis gives an overview of the legislation on cosmetics and description of the types of allergenic reactions caused by allergenic and potential allergenic metals in eye shadows. Listed below are the preparation methods of the samples for analysis and the list of the methods by which can the selected metals be analyzed. The experimental part of the diploma thesis deals with an analysis of selected allergenic and potentially allergenic metals in eye shadows. In the experimental part of diploma thesis method of sample preparation for analysis of eye shadows and a method of analysis of sample of eye shadows by ICP-MS have been optimized. Monitored analytes were selected metals. Altogether 6 samples of eye shadows from different manufacturers were chosen. Measured results have been statistically processed, confronted with the applicable legislation and assessed in terms of possible allergic reactions.
Analysis of selected allergens in fragrances

This thesis is focused on potentially allergenic substances (PASs) in cosmetics. Their specific features, methods of control and observation are described. There are also mentioned dermatology problems connected with usage of perfumed cosmetic products in which the allergenic ingredients can be found and possibilities of their regulation. Fragrances are mixtures of natural or synthetic character which give the aroma. In nature, fragrances are produced mainly by plants. More than one hundred fragrances have been identified as allergens. Twenty six of them were declared as PASs. Allergenic components of perfume compositions are the main cause of allergic contact dermatitis. Experimental part of the thesis is focused on the procedure of analysis of specific allergenic fragrances (?-Amylcinnamaldehyde, Anisealcohol, Benzylsalicylate, Cinnamaldehyde, ?-Hexylcinnamaldehyde, 7-Hydroxycitronellal and Methyl-2-octynoate). Headspace-Solid Phase Microextraction connected with Gas Chromatography-Flame Ionisation Detector (HS-SPME-GC-FID) was used for extraction, analysis and identification of these substances. The method was validated and applied on the spectrum of real samples of cosmetic products.
Analysis of aroma compounds in cosmetics by gas chromatography

This thesis is focused on aroma compounds which are presented in a wide variety of products including perfume, cologne, cosmetics, toiletries, laundry products and detergens. The widespread use and exposure to aroma compounds of aforementioned products could cause a range of the adverse reactions such as contact dermatitis, asthma, eczema and breathing problems. There are a lot of aroma compounds triggering negative allergic reactions but EU regulations limit 26 the most important fragrance allergens. The presence of these substances has to be declared on the product label if a limit of 0,01 % for rinse off and 0,001 % for leave-on products is exceeded. Of these 26 substances, a methodology was elaborated for determination of 12 allergens in a experimental part of this thesis. A method by solid-phase microextraction (SPME) and gas chromatography-flame ionization detection (GC-FID) was applied for fragrance allergens identification, extraction and measurement. The optimal extraction conditions of SPME-GC method were investigated including equilibrium time, extraction time and extraction temperature to increase extraction efficiency. Another important point is the method by SPME-GC-FID was validated and following performance parameters were determined: repeatability, linearity, limit of detection (LOD) limit of quantitation (LOQ). SPME-GC-FID was tested and applied to real samples.
Monitoring of allergenic aroma compounds in foods

This thesis deals with the identification and quantification of aromatic potentially allergenic substances which normally occurs in food, but their amount is not legislatively controlled. These aroma compounds have many positive and negative effects on human body, although they are not main types of allergens, several studies have highlighted their potentially allergenic character. For monitoring of the content of these substances non-chocolate confectionery were chosen as model samples. Theoretical part summarizes the production technology of non-chocolate confectionery from raw materials to final processing. It also deals with the characterization of 26 potentially allergenic aroma substances and their occurrence in nature and also in food or cosmetic products. In the experimental part potentially allergenic aroma compounds occurring in samples of jelly confectionery were identified and quantified using the SPME–GC–FID method.
