Conifers in the education of the biology

3 Abstract This thesis covers conifers (Pinopsida) and their use in teaching natural science in primary schools and biology in secondary schools. The aim is to analyze the problems of conifers (Pinopsida) at FEP (Framework Education Programme) for basic education and in selected textbooks, to determine the level of teaching conifers (Pinopsida) in primary schools and secondary schools in the Benešov region and propose a project with the theme of conifers (Pinopsida) for primary and secondary schools, with partial verification in the classroom. The theoretical part is concerned with conifers (Pinopsida) from a biological point of view. Characterize a group of conifers (Pinopsida), it's representatives in the Czech Republic and also deal with the ecology of conifers (Pinopsida). In the research part, using text and tables I mention results of analysis of FEP for basic education and analysis of textbooks. Later in the form of tables and graphs I mention the results of research on teaching conifers (Pinopsida) in primary and secondary schools in the Benešov region. Research was conducted using the questionnaire. Finally, I present the proposed project: The scent of resin partially verified in primary school. This work is intended for all who are interested in the conifers (Pinopsida) from the didactic point of view.
Somatic embryogenesis of conifers: description of structural development

Somatic embryogenesis is a very important tool of in vitro cultivation, therefore, it has recently devoted adequate attention. Stages of somatic embryogenesis involve several consecutive steps: induction of embryogenic culture and its proliferation, maturation of embryo, desiccation, germination and subsequent transfer of embryos into young seedlings. All of these steps are directly influenced by many factors, such as a choice of the original explant, time of cultivation, physical conditions or the composition of the culture medium. There are still emerging new studies describing how the culture conditions and, thus, the quality of embryos could be improved. An important prerequisite for further improving the quality of somatic embryos is good knowledge of aculture protocol, i.e. right concentrations of amended substances in the culture medium, the timing of each step, using the suitable explants etc. Another very important thing is good knowledge of development of all anatomical structures established during embryogenesis. Only then it will get closer to the point when somatic embryos will not arise only in the laboratory, but could be successfully used in practice. Analysis of primary photosynthetic processes in conifers: A comparison of selected methods and their possible utilisation for the study of genetic variability

Conifers are important both ecologically and socioeconomically, however, same parts of their biology are not that well researched. This includes genetics and breeding and partly even physiology. Because quantitative genetic analyzes applied in breeding necessitate an analysis of a large number of samples, and conventional methods of analysis are quite time-consuming, certain parameters describing e.g. the activity of photosynthetic electron-transport chain (ETC) are considered for such use. Several methods of the measurement of the activity of photosynthetic ETC exist, but there are some problems with their usage in conifers. I studied this issue from different points of view in three parts of this thesis. 1) I compared the photosynthetic ETC activity in 8 species of conifers using chlorophyll (Chl) fluorescence measurements on intact needles and polarographic measurements in isolated chloroplasts. Each method brought different information. 2) I measured Chl fluorescence parameters, reflectance spectra and pigment content in 536 genetically defined trees of Pinus sylvestris L. Utilization of plant bioindicators in monitoring of environmental contamination

Environmental contamination may be observed by different analytical tools as well as by various types of bioindicators providing relevant information about the effects of toxic substances and their mixtures towards organisms in situ. The aim of this work is to provide a comprehensive overview of plant bioindicators and biomonitors suitable for biomonitoring of persistent organic pollutants (POPs) in natural ecosystems. For groups of organic pollutants such as PCBs, PCDDs / Fs, PBDEs, PAHs, PFCs, and OCPs, review of literature is provided including knowledge about different species of coniferous trees, angiospermous plants (including crops), mosses and lichens that can be used for monitoring. In this work, analysis examples of samples of pine needles, bark, moss and lichen tissue are quoted as biomonitor tools. An overview of plants for phytotoxicity and genotoxicity observation using pollen grains abortivity and online bioindication as an example on non-specific biomonitoring is also provided.
