Studies of cell signaling triggered by the TRH receptor

Activation of thyrotropin-releasing hormone receptor (TRH-R) signaling has an irreplaceable role in a number of cellular processes. Thyroliberin (TRH) plays an important role in the regulation of secretion of other hormones and there are also mentions of its possible antiapoptotic and neuroprotective effects. On the other hand, TRH is quickly degraded and also its other properties, such as cardiac and endocrine side effects and low lipophilicity, disadvantage the therapeutic applications of this hormone compared to its various analogues. Due to their effects on the central nervous systém, TRH and its analogues represent a potential possibility for the treatment of various neurological diseasses, including neurodegenerative disorders. The molecular mechanisms responsible for the beneficial effects of TRH and its analogues have not yet been fully elucidated. So far, little is known about the involvement of TRH, or its analogues, in the regulation of energy metabolism or impact on the cellular phosphoproteome. Influence of opioid receptor genetic polymorphism on drug addiction.

Opioid substances have been used by mankind for several millenia not only for their ability to abolish even very intensive states of pain, but also for their ability to stimulate the brain reward circuit. However, a strong addiction can be formed to opioids quickly. This thesis focuses on collecting knowledge about the most abundant polymorphisms of these genes, their physiological impact and potential influence on drug addiction. The role of membrane cholesterol in delta-opioid receptor signaling Correlation with plasma membrane structure

Study of HEK293 cells stably expressing fusion protein between delta opioid receptor (δ-OR) and pertussis toxin-insensitive mutant of Gi1α protein, δ-OR-Gi1α (Cys351 -Ile351 ), provided the following results. Decrease of plasma membrane cholesterol content (cholesterol depletion) induced by cyclic oligosaccharide β-cyclodextrin did not affect binding of specific δ-OR agonist, [3 H]DADLE. Neither the maximum number of binding sites nor the affinity of [3 H]DADLE binding was changed by cholesterol depletion. However, the ability of δ-OR to activate cognate trimeric G proteins was impaired. EC50 value of agonist-stimulated [35 S]GTPγS binding was an order of magnitude higher. This effect was observed in case of both control and pertussis toxin-treated cells. It means that cholesterol depletion markedly reduced the efficiency of functional coupling of δ-OR to endogenously expressed pertussis toxin-sensitive G proteins of Gi/Go family as well as covalently bound Gi1α (Cys351 -Ile351 ) protein. Unchanged plasma membrane cholesterol content is therefore important requirement for proper δ-OR function. Business plan for home care agency

My following thesis introduces the home care segment and points to its present and future role in the field of health and social care. The aim of this thesis is to create a proposal of a business plan for home care agency, which would provide patients with health care in their home. The first part of this thesis describes the theoretical basis this thesis is based on. It introduces a concept of strategic management including a description of a strategic analysis, the structure of a business plan and there are also described aspects contemplated at the opening of the business. The theoretical part also includes detailed introduction of the home care segment including its development and history. The practical part starts with research and analytical chapters. These chapters describe the survey executed among general practitioners and a complete strategic analysis, whose results are summarized in the SWOT analysis. The business plan for the home care agency is based on these findings. In conclusion, I assess whether it would be possible to implement this business plan and I estimate the company's prospects.
