Use of selected fluorescence techniques to study of yeasts and yeast metabolites

The scope of thesis was the optimization of methods for the study of yeast and their metabolites using flow cytometry and fluorescence microscopy. Red yeasts are characterized by overproduction of carotenoids and lipids, which are used in food, pharmaceutical and feed industries. Currently, intensive research is being carried on to find appropriate microbiological alternatives for synthesis of these substances. Present thesis is focused on selected yeast genera: Rhodotorula, Sporobolomyces, Cystofilobasidium and strain Phaffia rhodozyma. Yeasts were cultivated on different nutrient media, in which glucose was used as a nutritional source, and also on glycerol and whey as waste material. In two strains - Cystofilobasidium macerans and Rhodotorula mucilaginosa growth characteristics were determined on a synthetic glucose production medium. All studied strains were able to use waste substrates as a source of nutrients. Some of the strains displayed increased production of carotenoids, and, additionally, in some cases also relatively high production of lipids. In classical cultivation in lipid and glucose medium supplemented with vitamins the best production characteristics displayed Rhodotorula glutinisstrain. In glycerol medium the highest amount of carotenoids and lipidic substances produced Sporobolomyces shibatanus strain. Strain Sporobolomyces roseus showed the best production characteristics on whey as the main source of carbon. The results show use of whey and glycerol seems like appropriate option for potential carbon source to cultivate carotenogenic yeasts and production of carotenoids and selected lipidic substances as products with higher added value. Further optimization of nutrient medium on the given substrates is needed for higher production of selected metabolites. Fluorescence microscopy and flow cytometry have proved to be suitable options for determination of the observed metabolites in the cells, their amount and viability.
Characterization of intermediates and waste products of some fermentation productions

This bachcelor thesis was aimed on characterisation of intermediate and waste products of some fermentation productions. The theoretical part of this thesis is aiming on characterization of raw material and its fermentation, process of distillation, waste product and introduction of main active components of all these intermediates. In the experimental part were characterized intermediates and products regarding the contents of sugar, ethanol and polyphenol. All these extracts contain particular levels of polyphenol and sugar corresponding to individual phases of fruit distillates production. The chemical characteristics of extracts from different phases of fruit distillates production were compared. Parameters as total phenolics, flavonoids, glucose, fructose and ethanol were evaluated. Subsequently the changes between individual phases were described. In conclusion, a solution of further use of waste products formed during fruit fermentation process was suggested.
Bioactive substances of rose hip

The aim of this master´s thesis was to determine the concentration of bioactive substances such as total polyphenols and flavonoids as well as the total antioxidant capacity in tea extracts from Rosa canina. In the second part, an experiment was found to detect antimicrobial activity and an effective concentration of ethanol against selected bacteria. The theoretical part is focused on description of natural antioxidants and their effect on free radicals, description of tea, description of rose hazel and possibilities of spectrophotometric determination of natural substances and eventually basic distribution of bacteria. In the experimental part, ethanol and aqueous extracts were prepared in which the total number of polyphenols, flavonoids and antioxidant effects. The results of the determination of polyphenols and flavonoids show that the solvent with ethanol is more suitable for flower fermentation and water is more suitable for dropping the rose. Comparison of the antioxidant activity of macerates showed no significant differences; slightly higher antioxidant activity showed macerates from pink flowers. The extracts showed antimicrobial activity against Micrococcus luteus and slightly against Bacillus cereus. The highest antimicrobial activity was determined for the micrococcus luteus
Preparation and characterization of food and cosmetic products containing selected vitamins

This bachelor thesis deals with the preparation and characterization of encapsulated forms of lipophilic vitamins in model conditions, focusing on cosmetic and food applications. The theoretical part describes the individual vitamins, their physiological effects, physical and chemical properties, sources and sequels of hypo/hypervitaminoses. Furthermore, their contribution in medicine, food industry and cosmetics, methods of determination and in the last part of the encapsulation method are discussed. Special attention is dedicated to the technique of particles encapsulation using liposomes, which was used in the practical part. The experimental part is focused on the study of liposomes of selected substances of vitamin nature. Namely, the issue of cholecalciferol, tocopherol and -carotene was addressed, for which a suitable method of determination was introduced. Subsequently, encapsulation of these substances and characterization of the prepared liposomes took place. Their size, stability and encapsulation efficiency, which reached high percentages, were determined. Liposomes were further put to the stability tests in digestive juices and model medium simulating cosmetic products. During the measured time, the release of encapsulated substances into the surroundings and their gradual degradation was monitored. At the end of the work, analytes were extracted from real foods and cosmetic products and the measured concentrations were compared with the declared values.
Characterization and application of some plant antimicrobial substances

The main goal of this bachelor thesis was the study of antioxidant and antibacterial effects of natural plant extracts and liposomes. The theoretical part was focused on description of natural antimicrobial and antioxidant substances from selected herbs and spices. Also bacterial resistance and methods used in work were described. In the experimental part basic characteristics of extracts from 12 kinds of herbs and spices (oil, alcohol and aqueous) were analyzed. Mainly content of polyphenols, flavonoids and the total antioxidant activity were determined. All extracts were also encapsulated into liposomes and the encapsulation effectivity, long-term stability and size and stability of liposomes were determined. The size of prepared liposomes was in range of 100 nm to 200 nm. Stability of the liposome particles was measured using a zeta potential and all prepared particles exhibited very good stability, but in term of long term stability were liposomes relatively unstable. Antibacterial activity was tested against Gram-positive Micrococcus luteus and Gram-negative Serratia marcescens. Encapsulation of extracts into liposome particles has greatly increased their antimicrobial effect and liposomes containing oil extracts from the goat, nutmeg and pepper and liposomes containing alcoholic extracts from star anise and cloves were the most effective against both strains. The prepared liposomes could be used as alternative to antibiotics and could be found application in the form of various antimicrobial gels for the cosmetic or pharmaceutical industry.
Uses of active compounds from honey in cosmetic products

The bachelor thesis was focused on the preparation and characterization of extracts and liposomes with active substances from chosen types of honey with the aim to apply these extracts to the cosmetic products. The extraction of active substances from chosen material was optimized during the work. As the most suitable organic solvent for extraction was chosen diethyl ether. All the extracts were tested on the content of total polyphenols, sugars and antioxidation activity. The final samples were also tested on the content of flavonoids. Extracts were encapsulated to the liposomes, which were tested especially for their antioxidation activity, size and stability. Then, extracts and liposome particles were applied to the cosmetic products. Cosmetic creams and gels were prepared and tested for antioxidation activity and antimicrobial activity against the strains Serattia marcescens and Micrococcus luteus. Antimicrobial activity of gels was proved especially against the strain of grampositive bacterias – Micrococcus luteus. From this point of view another one testing strain was chosen – Propionibacterium acnes. Finally, the chosen liposomes were tested for their contact with human cells, immortalized keratinocytes HaCat. No cytotoxic effect was observed and tested materials can be applied to cosmetics against acne.
Preparation of antimicrobial gels with plant extract and oils

This bachelor thesis was focused on studying antimicrobial effects of natural plant extracts and their subsequent application. The research was mainly focused on the antioxidant and antimicrobial activity of plant extracts and liposomal particles with their contents. Theoretical part of thesis was focused on the topic of natural antimicrobial and antioxidant substances, problems with bacterial resistance as well as describing features and attributes of used herbs, and characterization of gels and liposomes. Within the experimental part of thesis, three types of extracts (aquatic, ethanol and oil), prepared from 10 different types of plants, were described. The content of phenolic, flavonoid substances, also antioxidant and antimicrobial activity were then determined. After that, the contents of all extracts were encapsulated into liposomes, and size, stability, encapsulation activity, also antioxidant and antimicrobial activity of those liposomes were determined. The size distribution was wide, but in terms of the measured zeta potential, most of the particles were stable. All prepared extracts and liposomes containing them were tested for their antimicrobial activity. The tests were performed on a gram-positive genus of Micrococcus luteus and a gram-negative type of Serratia marcescens. The most effective oil extracts against both species were eucalyptus and fir, as well as oil extract of cloves, along with aqueous extracts of mustard seeds, cloves and juniper, or ethanol extract of anise which also has high antimicrobial effects. Finally, these extracts and liposomes were applied into the gels, which could be found useful in the pharmaceutical industry, as an alternative to externally used antibiotics, or in the cosmetic industry.
Encapsulation of active substances and possibilities of their application in anti-aging products

The aims of this thesis were encapsulation of selected active substances and their possible application in cosmetic products with anti-aging effects. 6 kinds of active substances were studied – lipophilic vitamin E and vitamin A, hydrophilic vitamin B1 and vitamin B2, and phenolic compounds ferulic acid and chlorogenic acid. In theoretocal part, the general characteristics of active compounds and their use are described. It also includes the description of used methods. Practical part was focused on characterisation of active compounds from the perspective of antioxidant activity and encapsulation into liposomes from soy and sunflower lecithin. Encapsulation effectivity, size, stability and long-term stability of liposomes after four weeks were determined. The most suitable liposomes with active compounds were also tested for safety on human skin cells using the MTT test of cytotoxicity on keratinocytes HaCaT. Furthernore, 3 types of cosmetic products were prepared – day cream, night cream and skin serum. Each kind of cosmetic product was devided into 4 types – the product without any active compounds, the product containing free active compounds, the product with liposomes with active compouds and the product containing empty liposomes. As active compounds all our tested substances were used. Final products were tested 14 days on 15 female volunteers. Before and after products usage parametres of their skin were measured to analyze effect of products on skin rejuvenation. Volunteers were also rating sensory analysis of cosmetic products. The part of this thesis was also creating an online questionnaire about anti-aging cosmetics for publicity to find out the popularity of this kind of cosmetics.
Production and characteritzation of extracellular hydrolases from selected moulds

This diploma thesis is focused on study of potential production of extracellular hydrolytic enzymes. The theoretical part deals with characterization of selected hydrolytic enzymes, their catalytic properties, the possibility of extracellular hydrolase production by fungi and their applications. In experimental part production strains Aureobasidium pullulans, Fusarium solani and Phanerochaete chrysosporium were used. Productions of cellulase, amylase, xylanase, lipase, protease and lignin-degraded enzymes (laccase, manganese- dependent peroxidase, lignin peroxidase) were observed. Cultivations were carried out in submersed mode in mineral medium supplemented by waste co-substrates such as wheat bran, corn bran, rice bran and oat bran, sawdust, rice, apple fiber, egg pasta and egg-free pasta. Production of enzymes depended on the substrate type and time of cultivation. The highest cellulase, xylanase and amylase activities were measured in the first period of cultivation (3 to 7 day). Lignin-degraded enzymes and proteases were produced at the end of cultivation (7 to 10 days). Lipolytic activity was detected only in A. pullulans, where the activity increased with time of cultivation. The highest value was determined during cultivation on wheat bran (3.6 nmol/ml.min). The highest xylanase and celulase activity (170.3 nmol/ml.min, 248.0 nmol/ml.min) were determined during cultivation of F. solani on corn bran. The highest amylase activity (111.8 nmol/ml.min) was reported in P. chrysosporium during the cultivation on rice. The highest protease activity (68.0 nmol/ml.min) was determined in F. solani grown on wheat bran. The best producer of laccase was A. pullulans, the highest production was recorded for egg-free pasta (27.0 nmol/ml.min). The maximum lignin peroxidase activity (12.5 nmol/ml.min) was measured during the cultivation of F. solani on egg pasta, while the highest yield of Mn-dependent peroxidase (7.7 nmol/ml.min) was achieved during the cultivation of A. pullulans on wheat bran. Lignin-degraded enzymes behaved as inductive, while the other enzymes were produced in mineral medium too. Activity of cellulase in the mineral medium was in A. pullulans strain higher than in media with waste substrates. Enzymes produced into A. pullulans medium were purified by ultrafiltration, ion exchange chromatography and gel filtration.
