This diploma theses focuses on the possibilities of metacognitive procedures use in learning and personal development of a student. It consists of theoretical bases of the topic as well as of empiric research of "minutes of learning", one of the metacognitive strategies which help students to adapt relevant learning abilities. In the theoretical part we can find the principles of education for 21st century and the key skills of a learner, as well as the fundamental curricular documents. In other chapters, the learning and personal development of a student is being focused. There are seven topics necessary for complex development of learner's personality. We get an overview of psychological concepts of the topics, and an insight of different psychological theories which are concerned with metacognitive strategies. The conditions for development of metacognitive dispositions of a learner are mentioned in the last chapter of the theoretical part. The methodological part states the aim of empiric research, describes the method used and defines the chosen sample. The third part - empiric research - examines the influence of the metacognitive strategy "minutes of learning" on the overall approach of a learner towards their own learning process. The empiric part consists of the analysis of research material, ways...

Hadravová, Jana ; Krčmářová, Tereza (advisor) ; Zemanová, Lenka (referee)
The Risk of Burnout in Primary School Teachers This diploma thesis deals with the risk of burnout syndrome in primary school teachers. The theoretical part of the diploma thesis provides an overview of the theoretical concept of burnout in teachers and the effects of burnout on their job performance and their personal life. The empirical part of the thesis introduces a qualitative research of the risk of burnout in primary school teachers who study the combined form of the 5- year Master's study programme at the Faculty of Education of Charles University, while working full-time. I interviewed several teachers who have been at the greatest risk of burnout, based on a questionnaire survey, about the factors that affect their mental health, about their perceived form of support and about their coping skills. Up to two-thirds of teachers experience exhaustion during their studies. Chronic stress, which is at the highest level during last years of the 5-year study programme, negatively affects their well-being, their family life and their physical and mental health. Key protective factors are problem-focused coping skills and work-life balance. The supportive attitude of the primary school management, as well as the attitude of the Faculty of Education towards studying teachers, can be also of...

Zemanová, Lenka ; Wildová, Radka (advisor) ; Šmelová, Eva (referee) ; Vykoukalová, Věra (referee)
Early reading literacy development of early readers at the beginning of the school attendance Abstract The aim of the dissertation is to monitor the development of the initial reading literacy of early readers, i.e., children who have learned to read with the comprehension before beginning of their school attendance. The aim of the research was to find answers to the questions how and when these children learned to read and how their initial reading literacy and motivation to read developed during the first year of primary school. Subsequently, the subject of the research was the verification of the connection between early reading and intellectual giftedness and family background. The theoretical part is devoted to current trends in primary school, the basic principles of primary education are described with emphasis on personal and constructivist conception of teaching, focusing on the topics of pupil assessment and inclusive education, including the education of gifted and exceptionally gifted students. The term reading literacy is defined, the stages of its development are distinguished and questions concerning reading are discussed, including the results of research on early reading in the Czech and foreign contexts. The chapter devoted to giftedness defines the term of giftedness and its...
Plasmid constuction for expression of selected reductase from Haemonchus contortus
Rychlá, Nikola ; Matoušková, Petra (advisor) ; Zemanová, Lucie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Nikola Rychla Supervisor: Doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Plasmid constuction for expression of selected reductase from Haemonchus contortus Barber's pole worm (Haemonchus contortus) is a pathogenic, gastroinstetinal, blood- sucking nematode. Drug resistance in H. contortus, and the associated haemonchosis of small ruminants such as sheep and goats, remain a major problem, and their biological and economic impacts are extensive. Given the fact that anthelmintics are still the gold standard of treatment, their reaserch is crucial, not only for farm animals but also for people. Carbonyl-reducing enzymes, such as aldoketoreductases and short-chain dehydrogenases/reductases, belong to the enzymes of the first phase of xenobiotics biotransformation and thus participate in drug metabolism and their increased elimination. Increased elimination leads to decreased toxicity in H. contortus and reduced efficacy of drugs in ruminants. Research of these enzymes can help tackle with drug resistance in H. contortus and contribute to the development of new drugs in the future. The aim of this diploma thesis is based on the construction of plasmid, which will be used for...
Study of unsteady phenomena in pump as turbine
Hliboký, Samuel ; Zemanová, Lucie (referee) ; Štefan, David (advisor)
This diploma thesis deals with centrifugal pump running as a turbine. Basic working principles of a pump are included, both in pump and turbine regime. Experimental data obtained from laboratory test bed are compared with CFD simulation on slightly simplified geometry. Obtained results are then processed using spectrogram. Influence of time step and mesh size on results is also researched.
Effect of monepantel on expression of biotransformation enzymes in sheep
Štolcová, Tereza ; Skálová, Lenka (advisor) ; Zemanová, Lucie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Tereza Štolcová Supervisor:prof. RNDr. Lenka Skálová Ph.D. Title of diploma thesis: Effect of monepantel on expression of biotransformation enzymes in sheep Monepantel (MOP) represents a new anthelmintic drug from the amino-acetonitrile derivatives group with the different mode of action in comparison with commonly used broad-spectrum drugs. It is used for the treatment and control of gastrointestinal nematode infection in sheep and cattle. This study focuses on the effect of MOP on expression of selected cytochromes P450 (CYP) in sheep in vivo and in vitro. The sheep treated with the recommended therapeutic dose of MOP was used for the in vivo experiment. Non-treated animals represented the control group. Changes of mRNA expression of selected CYPs in primary cultures of ovine hepatocytes were observed after 24 h incubation with MOP (10 μM) in the in vitro experiment. The qPCR method (real time quantitative PCR) was used for the quantification of transcription levels of mRNA. Both in vivo as well as in vitro results showed a significant up-regulation in CYP3A24 expression. The induction effect of MOP was even stronger than induction effect of well-known CYP3A inducer rifampicin....
Effect of the subchronic and early-life permethrin exposures on rat liver: oxidative stress and endogenous antioxidant responses
Palan, Matyáš ; Zemanová, Lucie (advisor) ; Dršata, Jaroslav (referee)
Charles University in Prague University of Camerino Faculty of Pharmacy in Hradec Králové Faculty of Pharmacy Department of Biochemical Sciences Department of Molecular Biology Candidate: Matyáš Palan Supervisor: Associate Prof. of Biochemistry Rosita Gabbianelli, Ph.D. RNDr. Lucie Škarydová, Ph.D. Title of diploma thesis: Effect of the subchronic and early-life permethrin exposures on rat liver: oxidative stress and endogenous antioxidant responses Permethrin is a widely used insecticide with a low acute toxicity to mammals; however, there are concerns about consequences of a long term and an early life exposure to this compound. Two different animal models were chosen in order to study effects of permethrin on liver. In the model of subchronic treatment, adolescent rats treated for 60 days with permethrin (150 mg/kg body weight/day) were used and sacrificed immediately after the treatment. As far as the model of early life is concerned, the treatment with permethrin (34.05 mg/kg body weight/day) lasted for 15 consecutive days in early life and sacrifice took place after the interval of 300 days since the beginning of the treatment. To evaluate the impact of permethrin treatment on liver tissue, markers of oxidative stress (lipid peroxidation, content of carbonyl groups), amount of superoxide...

CFD simulation of centrifugal pump working as a turbine (PaT) - influence of impeller side wall gaps on simulation results
Tomšej, Radek ; Zemanová, Lucie (referee) ; Štefan, David (advisor)
This master‘s thesis deals with a centrifugal pump running in reverse (turbine) mode, often called “pump-as-turbine”. The introduction lists basic principles and information about the direct (pump) and reverse modes. Several methodologies for predicting reverse operating point and efficiency evolution are used and compared with simulation results. Using CFD simulations, results obtained with simplified (excluding impeller side wall gaps) and complete geometry (including impeller side wall gaps) are assessed. Furthermore, the results of impeller trimming are evaluated with an aim to find the maximum power and efficiency. Impeller side wall gaps effect is analyzed in terms of friction torque and power loss.
