Vášková, Lucie ; Svobodová, Hana (advisor) ; Skálová, Lenka (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Lucie Vášková Supervisor: PharmDr. Hana Bártíková, PhD. Title of thesis: Changes of gene and protein expression of selected enzymes in mice due to cranberry antioxidants and obesity Obesity is a chronic disease coming up to endemic size in contemporary population. It is often associated with an adverse pathophysiological effects in the organism. This condition affects many processes occurring in the body and probably also activity and expression of antioxidant and biotransformation enzymes. Expression of antioxidant enzymes is influenced by dietary intake of antioxidants, which can be definitely beneficial to health. This thesis concerns about changes in protein and gene expressions of biotransformation and antioxidant enzymes by testing the four groups of experimental mice. This determination was carried out in normal and obese mice in which obesity was induced artificially by administration of monosodium glutamate. These individuals served as control group to the other two groups, which were used for testing the effect of cranberry extract on enzyme expression. The expressions were determined in subcellular fractions of livers and small intestine. The methods to determination of...
The impact of selected sesquiterpenes on the effectiveness of anticancer therapy in MCF-7 cell line
Vogričová, Lenka ; Svobodová, Hana (advisor) ; Hanušová, Veronika (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Title, Name, Surname of candidate: Lenka Vogričová Title, Name, Surname of tutor: PharmDr. Hana Bártíková, Ph.D. Title of a diploma work: The influence of selected sesquiterpenes on the effect of cytostatics in the MCF-7 cell line Sesquiterpenes are natural substances with numerous biological effects. Anti-tumour effect of certain sesquiterpenes has been determined. Doxorubicin is an antracycline- based medicine used for cancer chemotherapy, among other things, for the treatment of breast cancer. The doxorubicin therapy is limited by a number of adverse effects, particularly its cardiotoxicity. A combination of doxorubicin with suitable substances might result in lower therapeutic dosages of doxorubicin. One of the alternatives is the utilization of sesquiterpenes combined with doxorubicin. The purpose of this thesis is the determination of the effect of two sesquiterpenes, α-humulene and trans-nerolidol on the MCF-7 breast cancer cell line and the determination of the effect of α-humulene and trans-nerolidol on the effect of the doxorubicin cytostatic. The effect of α-humulene, trans-nerolidol, doxorubicin and a combination thereof on cell proliferation has been evaluated by the neutral red uptake...
Differences between susceptible and resistant strains of Barber's pole worm (Haemonchus contortus)
Bartásková, Lenka ; Svobodová, Hana (advisor) ; Lecová, Lenka (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Departement of Biochemical Sciences Candidate: Lenka Bartásková Supervisor: Mgr. Hana Bártíková, Ph.D. Title of the thesis: Differences between susceptible and resistant strains of Barber's pole worm (Haemonchus contortus) Haemonchus contortus is very common parasite. It can infect a large number of ruminant species and is a very dangerous parasite of sheep and goats worldwide. The infection caused by these worms is called haemonchosis. The symptoms are anaemia, loss of weight and death of the infected sheep and goats. Nowadays we can prevent this disease. The prevention of haemonchosis is available in the form of anthelmintic drugs. The most commonly administered drugs from this category are represented by benzimidazoles. However, the frequent use of anthelmintic leads to development of drug resistance in parasites. The resistance to anthelminthic drugs is caused by mutation at codon 200 of β-tubulin. The worms Haemonchus contortus can be divided into sub-groups according to their anthelmintic resistance. This thesis compares the number of alleles responsible for resistance in sensitive and resistant strains of Haemonchus contortus. A low percentage of the resistant allele is found in the sensitive strain ISE. In the resistant...

Kvita, Vojtěch ; Svobodová, Hana (advisor) ; Matoušková, Petra (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Vojtěch Kvita Supervisor: PharmDr. Hana Bártíková, Ph.D. Title of diploma thesis: The effect of green tea polyphenols on antioxidant enzymes in mice in vivo Antioxidant enzymes belong to compounds with proven effects on human organism. Their main purpose is to defend cells against oxidative stress. In order to support this process, we are taking substances called antioxidants either in regular diet or as dietary supplements. Recently there are a lot of companies on the market, offering the supplement, promising wight loss or defence against cancer. Green tea contains many natural substances, such as polyphenolic compounds which include catechins, which have proven effects on our bodies, but the specific way they act on selected antioxidant enzymes is still matter of study. The aim of this work was to determine the effect of green tea polyphenols on antioxidant enzymes in mice. We studied 8 groups (4 normal, obese 4) wherein the two groups were left as a control which means without administration of Polyphenon. Three groups were given a Polyphenon enriched diet and for various period of time (28 days, 3 days) and at different concentrations (0.1%, 0.01%). Activity was studied in liver...

Vášková, Lucie ; Svobodová, Hana (advisor) ; Hanušová, Veronika (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Lucie Vášková Supervisor: PharmDr. Hana Bártíková, PhD. Title of diploma thesis: The impact of cranberry extract and obesity on antioxidant enzymes in mice in vivo The obesity is the worldwide problem nowadays which threatens health of millions of people. The obesity is characterized as an excessive increase in the content of body adipose tissue. Increased quantity of adipocytes produces oxidized LDL particles which are deleterious for the body and damage the tissue of the organism. Fruit of Vaccinium macrocarpon contains range of natural substances. Proven effects of cranberries include action on the urinary tract infections, anti-atherogenic effects in the cardiovascular system, anticancer, antiulcer effect and also acts againts the development of the obesity. Most of these effects are largely determined by antioxidant actions of contained substances, particularly anthocyanidins. Recently, the nutritional supplements containing cranberries which are used frequently as a substitute for a varied diet or weight reduction have became very popular. The first aim of my study was to determine the influence of obesity on selected antioxidant enzymes of mice in vivo. Obesity by monosodium...
