Voltammetric Determination of Genotoxic 2-Nitrofluorene at Screen-Printed Carbon Electrodes
Jelínková, Lenka ; Vyskočil, Vlastimil (advisor) ; Zima, Jiří (referee)
of the Bachelor Thesis Voltammetric behavior of genotoxic 2-nitrofluorene (NF) has been investigated using direct current voltammetry (DCV) and differential pulse voltammetry (DPV) at a screen- printed carbon electrode (SPCE). The optimum conditions have been found for its determination in a concentration range from 1×10−6 to 1×10−5 mol l−1 for DCV technique and from 1×10−7 to 1×10−5 mol l−1 for DPV technique. An optimum medium for DCV at the SPCE was a 0.1 mol l−1 acetate buffer of pH 4.6, an optimum time of stirring of the sample prior to the analysis was 120 s (the attained limit of quantification (LQ) was 8.5×10−6 mol l−1 of NF). For DPV at the SPCE, the optimum medium was an acidic component part of the Britton - Robinson buffer of pH 2.0; the attained LQs were 6.2×10−7 mol l−1 of NF for the concentration range from 1×10−6 to 1×10−5 mol l−1 (at the stirring time of the sample of 120 s) and 2.0×10−7 mol l−1 of NF for the concentration range from 1×10−7 to 1×10−6 mol l−1 (at the stirring time of the sample of 300 s).
The role of innate immunity cells in the pathogenesis of celiac disease
Dáňová, Klára ; Palová Jelínková, Lenka (advisor) ; Černý, Jan (referee)
Celiac disease is an autoimmune disease which occurs in susceptible individuals after ingestion of food containing gluten. Gluten and its monomeric fraction gliadin induce inflammatory damage of the small intestine by activating the immune cells that react strongly to gluten peptides. Gluten peptides have the ability to activate cells of adaptive as well as innate immune system. This work is focused on the production of interleukin (IL)-1 in antigen presenting cells stimulated with peptic gliadin digest. We found that monocytes and peripheral blood mononuclear cells (PBMC) isolated from blood of celiac patients secrete significantly more IL-1α and IL-1β than cells of healthy donors after stimulation with gliadin digest. The gliadin-induced IL-1β expression is controlled by a signaling cascade that includes MAPK kinase family molecules and transcription factor NF-κB. Moreover, we found that the adaptor proteins MyD88 and TRIF as well as Toll-like receptor (TLR) 2 and 4 play a role in the signaling cascade underlying gliadin-induced IL-1β expression by using murine bone marrow derived dendritic cells (BMDC). The precursor form of IL-1β in gliadin- stimulated PBMC and murine BMDC is maturated by caspase-1. In celiac PBMC the gliadin- induced maturation and secretion of IL-1β depends on the potassium...
Problems of description of electronic online resources in contemporary library cataloguing practice
Jelínková, Lenka ; Bratková, Eva (advisor) ; Drobíková, Barbora (referee)
The topic of the thesis is bibliographic description of electronic online resources in foreign and Czech contemporary cataloguing practice. The first part of the thesis analyzes the development of description of electronic online resources in 90th of the 20th century along with an introduction of important foreign projects which contributed to this development. The paper analyzes in detail all areas of bibliographic description according to ISBD(ER) and the fields of MARC 21 format used for description of electronic online resources. The thesis also evaluates cataloguing practice of the National library of the Czech Republic within WebArchiv project and building of subject gateways. The heart of the thesis is detailed analyze of illustrating examples of description of electronic online resources including cataloguing records with comments clarifying some difficult questions of desciption of this resources. Final part of the thesis analyzes alternative approaches in the field of bibliographic description of electronic online resources, escpecially metadata formats and their most widespread representative within library community - Dublin Core. The thesis deals also with possible future perspectives of cataloguing practice and shortly introduces new cataloguing standard RDA.

Political business cycle and public procurements at regional authorities
Jelínková, Lenka ; Pavel, Jan (advisor) ; Holubářová, Jana (referee)
This bachelor thesis examines the political business cycle at public procurements, which were submitted by 13 regional authorities in the Czech Republic from September 2004 till June 2010. Two hypotheses are tested here. According the first one, the incumbents try to increase the probability of re-election by visible investments, e.g. public procurements for buildings. The second hypothesis supposes that the incumbents are afraid of not being re-elected. Therefore they strive for maximizing their personal profit from the office. Regression analysis is used for testing. Public procurements for buildings are tested separately from public procurements for goods and services. The number of public procurements, total amount and the average number of bidders are tested for each group of public procurements. The result is a detection of different motives in different parts of political business cycle.
