The social perception of social work studies by Charles University students
Vaněčková, Kateřina ; Novák, Petr (advisor) ; Cimbál, Václav (referee)
This master's thesis focuses on the study of social perception of the field of social work among students of Charles University who do not study this field. The thesis is structured into two main parts: theoretical and empirical. The theoretical part is devoted to defining and describing basic concepts. The empirical part contains a description of the used methodology - qualitative research and analysis of data obtained by a questionnaire survey. The main objective of the thesis was to find out how students of Charles University, who do not study social work, perceive this field in their social context. Another objective was to identify possible differences in the perception of this field between students of humanities and natural sciences disciplines. The research within this thesis has demonstrated that students of Charles University predominantly perceive the field of social work as highly beneficial to society. However, they also perceive it as strongly non-prestigious mainly due to its low financial remuneration and its demanding nature in terms of psychological aspects. A significant difference in the perception of this field between students of humanities and natural sciences disciplines was not proven.

Integration of people with serious mental illness into the open labour market
Syrovátková, Adéla ; Novák, Petr (advisor) ; Cooke, Dana (referee)
Diploma thesis "Integration of People with Mental Illness into the Open Labour Market" discusses the possibilities and conditions of employment in the open labour market for people with schizophrenia in the Czech Republic (focus on Prague 11+4). The paper first introduces the concept of "mental illness" from a theoretical point of view. In my work I first introduce this illness theoretically, I also use professional information from my practice and especially from my more experienced colleagues. I believe that drawing on the perspective of my clients and that of the professional staff is the most appropriate, as they have direct experience of it. In this thesis I conducted qualitative research, interviewing seven people who suffer from mental illness with whom the service has an established relationship. In the form of a questionnaire and a structured interview, I asked the patients, for example, how they perceive their life with mental illness, what difficulties they face in their life and how they manage to integrate into a "healthy" society, especially into employment on the open labour market.
The proces of acquiring social competences for people with hearing impairment
Mihočová, Jiřina ; Novák, Petr (advisor) ; Ondrušová, Jiřina (referee)
The presented diploma thesis analyzes how the process of acquiring social competences for people with hearing impairment takes place. The work is divided into theoretical and empirical parts and divided into four chapters. In the theoretical part, there are chapters clarifying the importance of hearing in the process of social competences and the effect of its lack on an individual's life. We will also find chapters clarifying the problem of hearing impairment and the definition of social competences with regard to hearing impairment. In the practical part, the research reveals the subjective attitude to which extent certain institutions were involved in the process in the life of the respondents. The practical part further analyzes whether the necessary social competences are the same from the point of view of the hearing and deaf communities. The mentioned research indirectly follows on from a part of the research that was described in the bachelor's thesis (Mihočová, 2022), where the research revealed what social competences are suitable for satisfactory social functioning from the perspective of hearing individuals. It was also conducted using a qualitative method. In the practical part, I also analyze the results of both research investigations and compare the knowledge gained. Based on the...
Drug problem in Jiřice prison
Kuchařová Roziňáková, Nikola ; Čedík, Miloslav (advisor) ; Novák, Petr (referee)
6 Abstrakt v angličtině The diploma thesis describes the problems in the Jiřice prison. The aim of this thesis is to introduce the readers to the issue of addictive substances in the prison and to introduce them to the issue of addictive substances in Jiřice prison. The first part of the thesis is devoted, with the use of relevant literature, to the description of the prison service of the Czech Republic, where the problems of the prison service, the profiling of prisons, the staffing of prisons, the legislation related to drug issues and the prison system are characterized. Furthermore, this part of the thesis describes the risks of substance consumption in the prisons, risk behaviour associated with substance consumption, what substances are used in prisons and the way of penetration of substances. In addition, the thesis includes a description of substance use prevention and addiction services provided in prison. The second part of the thesis deals with substance use issues in Jiřice Prison. Using the method of secondary data analysis and comparison, the thesis will examine how the substance abuse issues in Jiřice Prison have changed over the last five years. Firstly, each year is described separately and then it is compared how this issues has changed in the last five years. Specifically, the thesis...
The drug consumption rooms as one of the possible harm reduction tools?
Rosolová, Tereza ; Novák, Petr (advisor) ; Cooke, Dana (referee)
11 Abstract This thesis delves into a highly contentious subject that demands acknowledgment. Substance abuse is intricately linked with human existence, necessitating the recognition of this reality and the subsequent addressing of its repercussions. The topic of Drug Consumption Rooms (DCRs) as a viable harm reduction measure was selected based on both professional expertise and personal insights. The vicinity of the Anděl district in Prague 5, where I am actively involved, serves as a notable hub of drug activity in Prague. This is chiefly attributed to the shifting of the drug scene amid the COVID-19 pandemic. Merely two years ago, Prague 5 hosted two contact centers for drug users, resulting in a heightened concentration of substance users in the area. This surge correlated with an uptick in criminal activity and a decline in cleanliness and hygiene standards in the district. Among the potential remedies, the establishment of DCRs stands out. Despite its contentious nature, which has garnered attention from experts and the public alike, this idea has largely remained theoretical until recently. In September 2023, a pioneering pilot project of a mobile DCR was initiated in Brno, marking a significant breakthrough. It is my hope that this thesis will foster greater awareness of this issue, potentially...
Investigation of HSP70 oligomerization by structural mass spectrometry
Melikov, Aleksandr ; Novák, Petr (advisor) ; Jeřábek, Petr (referee)
Heat shock cognate protein 70 (HSC70) is a 71 kDa chaperone protein belonging to the ubiquitous family of heat shock proteins 70 (Hsp70). The representatives of this protein family are considered as molecular machines with ATP-hydrolase activity facilitating correct folding of spatial protein structure, both in normal and stressful conditions (hypoxia, heat shock, pH fluctuations etc.) In addition, HSC70 was identified as an uncoating enzyme for triskelion meshwork on the surface of clathrin-coated vesicles. Among other roles, HSC70 prevents protein aggregation and assists the polypeptide maturation, it facilitates the protein transport into organelles, such as endoplasmic reticulum and mitochondria. It is involved in targeting proteins for lysosomal degradation and in many other dramatically important cellular processes related to protein homeostasis. Therefore, the regulation of HSC70 and other HSP70 proteins is believed to be dramatically important, especially in a context of cellular stress. Based on the experimental observation, the mechanism of inactivation through oligomerization was hypothesized. The dimer and trimer species of Hsp70 proteins were identified both in case of prokaryotic and eukaryotic homologs. It was also speculated that Hsp40 cofactors promote oligomerization to even...
Utilization of pulse labelling techniques for studying the dynamics of proteins and protein complexes
Polák, Marek ; Novák, Petr (advisor) ; Obšil, Tomáš (referee) ; Vrbacký, Marek (referee)
(In English) Mass spectrometry (MS) techniques are routinely used to probe the structure and dynamics of proteins and protein complexes. Although MS techniques lack the high resolution of data provided by X-ray crystallography, NMR, or cryo-EM, they excel in providing insights into analyte dynamics, structure, and interactions with other components, such as ligands. This doctoral thesis presents a contribution to the field of structural biology employing and extending covalent labelling approaches, namely Fast Photochemical oxidation of Proteins (FPOP) and oxidation by singlet oxygen (1 O2). These approaches were followed to study the structure, dynamics, and interaction of proteins, nucleic acids, and protein-DNA complexes in solution. Initially, FPOP was used to investigate the interaction interface of FOXO4 and DAF16-DNA response element and to show the possibilities of analyzing such a complex using both 'bottom- up' and 'top-down' approaches. Furthermore, an isotope depletion strategy combined with multiCASI-ECD proved effective in delivering structural information with the highest possible resolution for mapping protein-DNA interfaces. This research showcases how information derived from structural proteomic methods can guide the construction of in-silico models for protein-DNA complexes with...
