Řád sekáči (Opiliones) představuje jednu z nejstarších linií pavoukovců s výskytem doloženým již z devonu. Navzdory své dlouhé evoluční historie, a osídlení široké škály suchozemských stanovišť, si sekáči zachovali relativně konzervativní morfologii. Tato morfologická uniformita však může skrývat vysokou skrytou druhovou diverzitu, kterou nelze spolehlivě odhalit pouze na základě zevních znaků. Cytogenetická analýza, zejména v kombinaci s morfologickými a molekulárně fylogenetickými výsledky, se tak stává potenciálním nástrojem pro odhalení skrytých rozdílů mezi jednotlivými liniemi. Předkládaná disertační práce se zaměřuje na podrobné studium karyotypů vybraných zástupců sekáčů s cílem prozkoumat jejich genetickou rozmanitost a evoluční dynamiku vybraných chromozomálních struktur. Výzkum je postaven na kombinaci klasických cytogenetických metod, které slouží k popisu počtu a morfologie chromozomů, a pokročilých molekulárně cytogenetických technik, zejména fluorescenční in situ hybridizace (FISH), prostřednictvím níž byla mapována lokalizace a variabilita klastrů 18S rDNA a vybraných mikrosatelitů. Zvláštní pozornost byla věnována odhadu možných chromozomových přestaveb, které mohly nastat během karyotypové diferenciace a hrají významnou roli při vzniku evolučních novinek v rámci jednotlivých... The order of harvestmen (Opiliones) represents one of the oldest lineages of arachnids, with occurrences documented as far back as the Devonian period. Despite their long evolutionary history and the colonization of a wide range of terrestrial habitats, harvestmen have maintained relatively conservative morphology. However, this morphological uniformity may conceal high hidden species diversity that cannot be reliably detected based solely on external features. Cytogenetic analysis, especially when combined with morphological and molecular phylogenetic results, thus becomes a potential tool for uncovering hidden differences between individual lineages. The presented thesis focuses on a detailed study of the karyotypes of selected representatives of harvestmen, aiming to explore their internal genetic diversity and the evolutionary dynamics of selected chromosomal structures. The research is based on a combination of classical cytogenetic methods, used to describe the number and morphology of chromosomes, and advanced molecular cytogenetic techniques, particularly fluorescence in situ hybridization (FISH), through which the localization and variability of 18S rDNA clusters and selected microsatellites were mapped.
