Dvoukomponentní signalizační systémy představují jeden z nejrozšířenějších způsobů signální transdukce u bakterií, který je využíván pro detekci podnětů z okolí a následné zprostředkování adekvátní odpovědi na přijatý signál. Jedním z modelových zástupců dvoukomponentních signalizačních systémů je proteinový komplex histidinkinázy obsahující globinovou doménu z bakterie Anaeromyxobacter sp Fw. 109-5 (AfGcHK) a příslušného regulátoru odpovědi. Histidinkináza AfGcHK slouží jako senzor kyslíku, který umožňuje bakterii určit, zda se nachází v aerobním či anaerobním prostředí. Aktivace AfGcHK spočívá v navázání kyslíku na molekulu hemu v senzorové doméně, které je následováno autofosforylační reakcí ve funkční doméně enzymu. Další krok zahrnuje fosfotransferázovou reakci, při které je fosforylová skupina z AfGcHK přenesena na příslušný regulátor odpovědi. Doposud nebyla objasněna struktura komplexu AfGcHK:RR přímými metodami, a také není zcela známý mechanismus fosfotransferázové reakce. Předmětem této práce je zmíněné odhalení struktury komplexu užitím kryoelektronové mikroskopie a následná diskuse možných způsobů přenosu fosforylové skupiny mezi jednotlivými komponentami komplexu. Klíčová slova: dvoukomponentní signalizační systémy, histidinkináza, regulátor odpovědi, kryoelektronová mikroskopie Two-component signalling systems represent one of the most widely used signal transduction pathways in bacteria, which are used to detect environmental stimuli and then mediate an appropriate response to the received signal. A model representative of two-component systems is the histidine kinase protein complex containing the globin domain from Anaeromyxobacter sp Fw. 109-5 (AfGcHK) and the corresponding response regulator. The AfGcHK histidine kinase serves as an oxygen sensor that allows the bacterium to determine whether it is in an aerobic or anaerobic environment. Activation of the AfGcHK histidine kinase involves the binding of an oxygen molecule to the sensor domain heme molecule, which is followed by an autophosphorylation reaction in the functional domain of the enzyme. The next step involves a phosphotransferase reaction involving the phosphoryl group from AfGcHK being transferred to the appropriate response regulator. To date, the structure of the AfGcHK:RR complex has not been elucidated by direct methods, and the mechanism of the phosphotransferase reaction is also not fully understood.
