The role of TRPV4 channels in disorders of central nervous system ABSTRACT Astrocytes are responsible for maintenance of the ionic and water homeostasis in the central nervous system (CNS), and they are associated with pathologies, such as Alzheimer's disease (AD) or ischemia. In this study, we investigated astrocyte volume regulation, which reflects astrocytic ability to buffer ions, neurotransmitters and water. We focused mainly on the role of Transient Receptor Potential Vanilloid 4 (TRPV4) and Aquaporin 4 (AQP4) channels that can contribute to the development of a brain edema during ischemic brain injury. We employed a model of focal cerebral ischemia (FCI) and magnetic resonance imaging to assess the extent of the damage, as well as in situ model of oxygen-glucose deprivation (OGD), real-time iontophoresis to assess the parameters of the extracellular space or three-dimensional (3D) morphometry to assess astrocyte volume changes. Additionally, we investigated changes in the expression of genes contributing to astrocyte volume regulation in the mouse model of AD as well as in mice with a deletion of TRPV4 and/or AQP4. Úloha TRPV4 kanálů v poškození centrálního nervového systému ABSTRAKT Hlavní funkcí astrocytů v centrální nervové soustavě (CNS) je udržování homeostázi vody a iontů, a jsou často spojovány s patologiemi, jako Alzheimerova demence (AD) nebo ischemické poškození. V této práci jsme se zaměřili na objemovou regulaci astrocytů, protože odráží jejich pufrovat ionty, neurotransmittery a vodu. Především jsme se soustředili na úlohu Transient Receptor Potential Vanilloid 4 (TRPV4) a Aquaporin 4 (AQP4) kanálů v ischemickém poškození mozku, během něhož mohou přispívat k rozvoji edému. Použili jsme model fokální cerebrální ischemie a magnetickou rezonanci ke zhodnocení rozsahu poškození, ale i in situ model kyslíkové-glukózové deprivace (OGD), iontoforetické stanovení parametrů extracelulárního prostoru a třírozměrnou (3D) morfometrii ke sledování objemových změn astrocytů. Současně jsme analyzovali změny v expresi genů, které se podílejí na regulaci objemu astrocytů, a to jak v myším modelu AD, tak i u myší s delecemi TRPV4 a/nebo AQP4. V modelu AD jsme identifikovali změny v expresi proteinů, které přispívají k transportu vody v mozku, včetně AQP4 a zároveň naše analýza ukázala sníženou schopnost astrocytů měnit svůj objem. Použití modelu OGD a myší s deletovaným TRPV4 a/nebo AQP4 nám umožnilo objasnit jejich úlohu...
