PharmDr. Martina Čečková, Ph.D. Název diplomové práce: Vliv nového FLT3 inhibitoru na indukci buněčné smrti u leukemických buněk Zhruba u 30 % pacientů s akutní myeloidní leukemií se vyskytuje mutace kinázy Flt3, která je jednoznačně negativním prognostickým faktorem nemoci. Proto vývoj nových Flt3 inhibitorů představuje zásadní pokrok v terapii AML u těchto pacientů. Ačkoli standardní léčba chemoterapeutiky (cytarabinem a antracyklinem) a alogenní transplantace kmenových buněk, které se využívají v terapii AML posledních 40 let, v mnoha případech vedou ke kompletní remisi, zároveň však nesnižují četnost relapsu a přináší mnoho rizik spojených s působením na zdravé buňky organismu. Účelem rozvoje cílené terapie, včetně inhibitorů Flt3 kinázy, je zabránit tomuto nežádoucímu působení chemoterapeutik. Cílem výzkumu prováděném v rámci vypracovávání této diplomové práce bylo stanovení vlivu nově syntetizovaného Flt3 inhibitoru K1872 na proliferaci leukemických buněk s wild type Flt3 kinázou (THP-1) a buněk s nejčastěji se vyskytující mutací Flt3-ITD (MOLM-13 a MV-4-11). Pomocí níže uvedených metod se zkoumal vliv inhibitoru na proliferaci buněk, navození apoptózy a... PharmDr. Martina Čečková, Ph.D. Title of diploma thesis: Effect of a new FLT3 inhibitor on induction of cell death in leukemia cell lines Approximately 30% of patients with acute myeloid leukemia have a Flt3 kinase mutation, which is clearly a negative prognostic factor of the disease. Therefore, the development of new Flt3 inhibitors represents a major advance in the therapy of AML in these patients. Although standard treatment with chemotherapeutics (cytarabine and anthracycline) and allogeneic stem cell transplantation, which have been used in AML therapy for the past 40 years, lead to complete remission in many cases, they do not reduce the frequency of relapse and bring many risks associated with the effect on healthy cells of the body. The purpose of developing targeted therapy, including Flt3 kinase inhibitors, is to prevent this adverse effect of chemotherapeutics.
