PharmDr. Vendula Hepnarová, Ph.D. Název diplomové práce: Hodnocení farmakologických vlastností nových potenciálních léčiv Alzheimerovy choroby Alzheimerova choroba je neurodegenerativní onemocnění s progresivním a ireverzibilním průběhem. Příčina této nemoci není doposud přesně známa. V současnosti jsou dostupná čtyři léčiva, která byla schválena Státním ústavem pro kontrolu léčiv. Tři z nich působí jako inhibitory cholinesteras (donepezil, rivastigmin, galantamin) a čtvrté jako antagonista N-methyl-D- aspartátových (NMDA) receptorů (memantin). Bohužel tato léčiva neovlivňují průběh nemoci a poskytují pouze dočasnou symptomatickou úlevu. Důležitou oblastí současného výzkumu jsou MTDLs (Multi-Target Directed Ligands), které působí na více cílových struktur souvisejících s patofyziologií nemoci. Tyto látky představují slibnou terapeutickou strategii. Cílem této práce bylo změřit schopnost inhibovat lidskou acetylcholinesterasu (AChE) a butyrylcholinesterasu (BuChE) u 58 nových potenciálních MTDLs vzniklých substitucí bifenylu a benzhydrylu s použitím Ellmanovy metody a zároveň u vybraných sloučenin spektorfotometricky... The cause of AD is still not exactly known. There are currently four drugs available that have been approved by the State Institute for Drug Control. Three of them act as cholinesterase inhibitors (donepezil, rivastigmine, galantamine) and the fourth one as a N-methyl-D-aspartate (NMDA) receptor antagonist (memantine). Unfortunately, these drugs do not affect the course of the disease and provide only temporary symptomatic relief. An important area of current research is MTDLs (Multi-Target Directed Ligands), which act on multiple target structures related to the pathophysiology of the disease. These agents represent a promising therapeutic strategy.
