Předložená bakalářská práce je zaměřena na studium vybraných vlivů na produkci biopolymerů, polyhydroxyalkanoátu (PHA) a extracelulárních polysacharidů (EPS), extrémně halofilní Archae Haloferax mediterranei. V teoretické části je zpracována stručná charakteristika biopolymerů a přehled příslušných mikrobiálních producentů. Experimentální část byla zaměřena na výběr vhodného substrátu, koncentraci soli a glukózy v médiu pro optimální produkci biopolymerů. Dalšími studovanými vlivy byla přítomnost světla a vhodná kultivační teplota. Pro stanovení biomasy byla zvolena metoda UV-VIS spektrofotometrie a gravimetrická metoda. PHA byly analyzovány pomoci plynové chromatografie s FID detekcí. Konkrétním PHA, produkovaným pomoci H. mediterranei, je kopolymer P(3HB-co-3HV). Obsah 3HV v kopolymeru se pohybuje okolo 10 %. Pro produkci PHA se jako nejlepší substrát osvědčila směs glukózy a glycerolu, kdy bylo dosaženo až 80,6% PHA v sušině. Dále byla optimalizována osmotická síla prostředí, kde se z pohledu akumulace PHA nejlépe osvědčila koncentrace NaCl 200 g/l. Je zajímavé, že na různou iontovou sílu prostředí reagovala kultura odlišnou pigmentací. Posledním klíčovým parametrem, který byl optimalizován v rámci této práce, byla teplota. Nejvyšší výtěžek PHA byl zaznamenán při teplotě 37°C, nicméně kultura byla schopna růst a akumulovat PHA i při teplotě 50°C. This bachelor thesis is focused on study of selected effects on production of both polyhydroxyalkanoates (PHA) and extracellular polysaccharides (EPS) by halophile Archae Haloferax mediterranei. Theoretical part deals with a brief characterization of biopolymers and provides overwiev of the relevant microbial producers. The experimental part is focused on selection of a suitable substrate, concentration of sodium chloride and glucose in culture medium for a optimal production of biopolymers. Effects of light and suitable cultivation temperature were studied as well. UV-VIS spectrophotometry and gravimetry were used for analysis of biomas content. The content of PHA was analysed by gas chromatography with FID detection. PHA produced by H. mediterranei is a copolymer P(3HB-co-3HV), 3HV content is about 10%. The mixture of glucose and glycerol proved to be the most suitable substrate for a production of PHA, with a content od 80,6% of PHA. Further, we investigated optimal concetration of NaCl, which was found at 200 g/l. Interestingly, microbial culture responded to various salt concertations by differences in pigment formation. Finally, the last parameter optimalized in this work was temperature of cultivation. The highest content of PHA was obtained at 37°C, but the culture was capable of PHA accumulation and growing also at 50°C.
