Tato práce popisuje vznik obrazu v transmisním světelném mikroskopu s proměnnou aperturou kondenzoru. Tento způsob pořizování obrazu je výhodný při pozorování tlustých vzorků, což jsou objekty, u kterých je v mikroskopickém měřítku významná výška. Klasické transmisní mikroskopy pro tento výzkum vhodné nejsou, protože výsledný obraz, který produkují, obsahuje patrné informace z velkého objemu vzorku mimo oblast, která je vyšetřovaná. Tento problém byl vyřešen po vynálezu konfokálního mikroskopu, který je ovšem daleko dražší a má i některé nevýhody navíc. Cílem je zpracovat obrazy pořízené pomocí kondenzoru s rotující aperturou tak, aby došlo k redukci podílu nežádoucí informace a obraz se tak stal "čistším". Tato metoda zpracování obrazu nemohla být použita v minulosti, protože tehdejší počítače neměly dostatečnou paměť a výkon. Na tuto metodu se dá nahlížet jako na převod množiny výstupů z mikroskopu s vylepšenou osvětlovací soustavou na odpovídající množinu výstupů konfokálního mikroskopu.
This thesis describes possible methods of processing images acquired using a transmitted light (aka. widefield) microscope with adjustable condenser aperture. This method is useful when examining thick specimen that have significant third dimension. Classical microscopy is not a very good method to research this type of specimen since the output of widefield microscope incorporates very much information from areas that are not of any interest. This is a serious complication in research of thick specimen (frequently seen in medicine and biology), so when a remedy appeared (confocal microscope), everybody started using it. However, these microscopes are very expensive and have even other drawbacks. The aim is to process images acquired by the method involving rotating condenser aperture using new computer-based techniques. These techniques were not available when scientists begun their research on thick specimen since computers were not so widespread and didn't have enough memory. This method of image processing can be considered as a software substitution of hardware that makes difference between classical and confocal microscopes. In some cases, it can produce exactly the same results.
