Barbora Boltnarová Posluchač: Natalie Wurzel Název diplomové práce: Příprava a hodnocení nanočástic pro cílenou léčbu zánětlivých onemocnění Nanočástice jsou ve zdravotnických vědách využívány kromě jiného pro cílené dodání léčiv. Toho se využívá pro terapii nádorových a chronických zánětlivých onemocnění. Cílem této diplomové práce bylo připravit nanočástice vhodné pro cílenou léčbu zánětlivých onemocnění. Úkolem bylo stanovit ideální koncentraci účinné látky, která zajistí vysokou enkapsulační efektivitu a drug loading. Zkoumal se také rozdíl mezi nanočásticemi připravenými z PLGA 5/5E a PLGA 7/3A. Nanočástice byly připraveny metodou nanoprecipitace. Matrici nanočástic tvořil PLGA s různým poměrem kyseliny mléčné a glykolové. Jako organické rozpouštědlo byl použitý aceton. Enkapsulovanou léčivou látkou byl protizánětlivě působící dexametazon acetát. Vodnou fázi tvořil 0,1% vodný roztok Pluronicu F-127. Mezi hodnocené parametry vzniklých nanočástic patřila velikost, index polydisperzity, zeta potenciál, enkapsulační efektivita a drug loading. This is used for the therapy of cancer and chronic inflammatory diseases. The aim of this diploma thesis was to prepare nanoparticles suitable for targeted treatment of inflammatory diseases. The task was to determine the ideal concentration of the active substance, which would ensure high encapsulation efficiency and drug loading. The difference between nanoparticles prepared from PLGA 5/5E and PLGA 7/3A was also investigated. Nanoparticles were prepared by the nanoprecipitation method. The matrix of the nanoparticles was composed of PLGA with different ratios of lactic acid and glycolic acid. Acetone was used as the organic solvent. The encapsulated active substance was the anti-inflammatory dexamethasone acetate. The aqueous phase was consisted of a 0.1% aqueous solution of Pluronic F-127.
