Molekuly MHC I. třídy (MHC I) hrají významnou roli v adaptivní imunitě, jelikož prezentují na buněčném povrchu antigenní peptidy pocházející z intracelulárního prostředí. Jsou rozeznávány CD8+ T-lymfocyty a mohou také trans-interagovat s receptory NK buněk. MHC I se skládají z těžkého řetězce, z β2 mikroglobulinu (β2m, lehký řetězec) a peptidu, což tvoří uzavřenou konformaci MHC I. β2m je nekovalentně připojen k těžkému řetězci sestávajícího se z extracelulární domény (podjednotky α1, α2, α3), transmembránové domény a cytoplazmatické domény (s obsahem konzervovaných motivů). Při aktivním buněčném metabolismu může dojít k disociaci peptidu a β2m z těžkého řetězce MHC I, a tím dochází k vytvoření otevřené konformace. Vlivem změny konformace dochází k rozbalení podjednotky α1, která tak může interagovat s různými receptory a molekulami. Otevřené MHC I vytvářejí cis-interakce buď sami se sebou, čímž vznikají homodimery realizujících imunologické funkce, nebo s receptory na buněčném povrchu tvořících heterodimery, které realizují neimunologické funkce. Mimo povrch buňky se vyskytují v rozpustné formě. Cis-asociace jsou velmi důležité, jelikož ovlivňují buněčnou signalizaci a inhibici/aktivaci T-lymfocytů a NK buněk či únik nádorových buněk před imunitou. Přesný mechanismus vzniku, exprese a funkce... The major role of MHC class I molecules in adaptive system is to present antigen peptides derived from intracellular environment on the cell surface. These peptides are recognized by CD8+ T-lymphocytes and they can also interact with NK cells via trans-interaction. MHC class I molecules are composed of a heavy chain, β2-microglobulin (β2m, light chain) and peptide, forming a closed conformation. The heavy chain is non-covalently associated with the light chain and is folded into extracellular domain (α1, α2, α3 subunits), transmembrane domain and cytoplasmic domain (with conserved motifs). Upon active metabolism, the β2m and peptide may dissociate from the MHC I heavy chain what leads to the formation of open conformations of MHC I. This conformational change causes the subunit to unfold and allow its interaction with various receptors and molecules. Open conformers of MHC I may form cis-interactions with themselves creating homodimers involved in immunological functions or they can associate with different receptors on the cell surface creating heterodimers responsible for non-immunological functions.
