Spolu s širokým využíváním herbicidů v moderním zemědělství se objevil fenomén herbicidní rezistence, kdy se plevelné rostliny stávají částečně nebo zcela odolnými vůči působení jednoho nebo více herbicidů. Rezistence vzniká dvěma způsoby - změnou v cílové molekule, na kterou herbicid působí, tzv. "target site resistance" (TSR), nebo změnou v jiných necílových proteinech, tzv. "non-target site resistance" (NTSR). Na NTSR se podílí UDP-glykosyltransferázy (UGT), které glykosylují široké spektrum endogenních rostlinných molekul a xenobiotik. Rovněž zvýšená aktivita antioxidačních enzymů může přispívat k NTSR. Tato práce byla zaměřena na interakci herbicidů pinoxaden a sulcotrion s modelovou rostlinou Arabidopsis thaliana. Cílem této práce bylo zjistit, zda některé z vybraných UGT genů mění svůj expresní profil po kontaktu s pinoxadenem, a zda má na tyto změny vliv, jestli se rostliny v předchozích generacích s pinoxadenem již setkaly. Exprese vybraných UGT genů byla také sledována u rostlin ošetřených sulcotrionem. Dále bylo sledováno, zda se u rostlin, které byly ošetřeny herbicidem pinoxaden nebo sulcotrion, mění aktivita vybraných antioxidačních enzymů. Výsledky ukázaly, že v případě rostlin A. thaliana, které přišly do kontaktu s herbicidy, došlo u některých UGT genů k signifikantní změně v... Along with the widespread use of herbicides in modern agriculture, the phenomenon of herbicide resistance has appeared, where weed plants become partially or completely resistant to the action of one or more herbicides. Resistance arises in two ways - by a change in the target molecule on which the herbicide acts, so-called "target site resistance" (TSR), or by a change in other non-target proteins, so-called "non-target site resistance" (NTSR). UDPglycosyltransferases (UGT), which glycosylate a wide range of endogenous plant molecules and xenobiotics, are also involved in NTSR. Also, increased activity of antioxidant enzymes may contribute to NTSR. This work was focused on the interaction of the herbicides pinoxaden and sulcotrione with the model plant Arabidopsis thaliana. The aim of this work was to determine whether some of the selected UGT genes change their expression profile after contact with pinoxaden, and whether these changes are influenced by the contact of plants in previous generations with pinoxaden.
