Kobalt je prvek, který prostřednictvím hypoxií indukovaných faktorů simuluje hypoxii za normoxických podmínek. Jedním z faktorů, které mohou vyvolat buněčnou senescenci, je hypoxie. Cílem práce bylo hodnocení proteinů p16INK4a a Ki67 jako markerů buněčné senescence u H9c2 buněčné linie po expozici CoCl2. Metodika: Byla změřena cytotoxicita pomocí MTT testu na buněčné linii H9c2 po 24 a 48hodinové expozici CoCl2. Následně byla pomocí RT-PCR metody zkoumána relativní genová exprese genů spojených s hypoxií (HIF1α a BNIP3) a senescencí (p16INK4a a Ki67) u H9c2 po expozici kobaltem. Výsledky: Expozice H9c2 buněk CoCl2 vyvolala dávkově a časově závislou toxicitu. Po 24hodinové expozici nebyla pozorována toxicita CoCl2 až do koncentrace 100 μM, hodnota IC50 byla 500,5 μM. Cobalt is an element that simulates hypoxia under normoxic conditions through hypoxia-inducible factors. One of the factors that can induce cellular senescence is hypoxia. The aim of the work was to evaluate p16INK4a and Ki67 proteins as markers of cell senescence in an H9c2 cell line after exposure to CoCl2. Methods: Cytotoxicity was measured using the MTT assay on the H9c2 cell line after 24 and 48 hours of exposure to CoCl2. Subsequently, relative gene expression of genes associated with hypoxia (HIF1α and BNIP3) and senescence (p16INK4a and Ki67) in H9c2 after exposure to cobalt was investigated using the RT-PCR method. Results: Exposure of H9c2 cells to CoCl2 induced dose- and time-dependent toxicity.
