[cze][eng] Obezita je velmi nebezpečným metabolickým onemocněním, vyvolaným rozvojem rezistence na anorexigenní (příjem potravy snižující) hormon leptin, s neustále narůstající prevalencí. Jako kandidáti pro vývoj antiobezitních léčiv se v poslední době ukazují jak anorexigenní peptidy, tak antagonisté orexigenních (příjem potravy zvyšujících) peptidů. Příkladem takového peptidu je nově objevený endogenní antagonista ghrelinu označovaný jako antimikrobiální peptid produkovaný játry 2 (LEAP2; z angl. liver enriched antimicrobial peptide 2). Jedinečnost vztahu mezi ghrelinem a LEAP2 spočívá v neobvyklosti výskytu agonisty a antagonisty stejného receptoru v rámci jednoho organismu. Tento receptor známý jako receptor pro sekretagogy růstového hormonu 1a (GHSR1a; z angl. growth hormone secretagogue receptor 1a) zodpovídá mimo jiné za zvýšení příjmu potravy a nárůst tělesné hmotnosti. Pro studium těchto peptidů byl použit přirozený ghrelin a analogy LEAP2, konkrétně nelipidizovaný fragment LEAP2(1-14) a lipidizovaný (palmitovaný) palm-LEAP2(1-14). Cílem této práce bylo in vitro prokázat vazbu analogů LEAP2 na receptor GHSR1a a porovnat efekt lipidizovaného analogu oproti nelipidizovanému. Účinky lipidizovaného analogu byly dále sledovány in vivo. Výsledky in vitro pokusů prokázaly vazbu obou analogů LEAP2 ke GHSR1a...Obesity is a very dangerous metabolic disease, caused by the development of resistance to the anorexigenic (food intake reducing) hormone leptin, with an ever-increasing prevalence. Recently, both anorexigenic peptides and orexigenic (food intake increasing) peptide antagonists have emerged as candidates for the development of anti-obesity treatment. An example of such a peptide is the newly discovered endogenous ghrelin antagonist known as liver enriched antimicrobial peptide 2 (LEAP2). The uniqueness of the relationship between ghrelin and LEAP2 lies in the unusual occurrence of agonists and antagonists of the same receptor within the same organism. This receptor, known as growth hormone secretagogue receptor 1a (GHSR1a), is, among other roles, responsible for increased food intake and weight gain. Natural ghrelin and LEAP2 analogues, specifically non-lipidized LEAP2(1-14) fragment and lipidized (palmitoylated) palm-LEAP2(1-14) were used for studying their properties. The aim of this study was to demonstrate binding properties of LEAP2 analogues to the GHSR1a and to compare the effect of the lipidized analogue versus the non- lipidized one in vitro.
