Alzheimerova choroba je nejrozšířenější chronické a progresivní neurodegenerativní onemocnění. Hlavní problém této choroby je poškození a snížení počtu nervových buněk. Tím dochází k úbytku mozkové hmoty a rozvíjí se poškození paměti. Onemocnění postihuje především lidi ve starším věku, u nichž bývá nejčastější příčinou demence. Celosvětově tímto onemocněním trpí zhruba 50 milionů lidí, přičemž jejich počet roste. Alzheimerova choroba tak představuje závažný problém veřejného zdraví. Patogeneze této choroby souvisí s produkcí a ukládáním peptidu amyloidu beta. Tento peptid má tendenci agregovat. Jeho nižší oligomerní formy jsou ve vodě rozpustné a působí toxicky na nervové buňky. Oligomery amyloidu beta tak představují možný cíl pro diagnostiku Alzheimerovy choroby. Oligomerní formy amyloidu beta mají nekovalentní charakter, což může negativně ovlivňovat jejich stabilitu při analýzách v modelových a biologických vzorcích. V této bakalářské práci byly hledány vhodné podmínky pro přípravu modelových vzorků amyloidu beta. Bylo pozorováno, jak se mění distribuce oligomerních forem působením rozpouštědel, solí a dalších aditiv. Byl také zkoumán vliv pH na inkubaci amyloidu beta v octanu amonném a ve vodě. Na závěr byla vyvinuta metoda pro analýzu amyloidu beta ve vzorcích mozkomíšního moku bez... Alzheimer's disease is the most widespread chronic and progressive neurodegenerative disease. The main problem of the disease is the reduction and damage of nerve cells. This leads to loss of brain mass and memory impairment. The disease mainly affects the elderly, for whom it is the most common cause of dementia. Around 50 million people suffer from this disease worldwide, and the number is growing. Alzheimer's disease thus represents a serious public health problem. The pathogenesis of this disease is controlled by the production and deposition of amyloid beta peptide. This peptide tends to aggregate. Its lower oligomeric forms are water-soluble and have a toxic effect on nerve cells. Amyloid beta oligomers therefore represent a possible target for the diagnosis of Alzheimer's disease. Oligomeric forms of beta amyloids have a non-covalent character, which can negatively affect their stability during analyzes in model and biological samples. In this bachelor thesis, suitable conditions for the preparation of model amyloid beta samples were sought. It was observed how the distribution of oligomeric forms changes under the influence of solvents, salts and other additives.
