Tato diplomová práce je zaměřena na přípravu kovalentně zesítěných oligomerních forem amyloidu beta a jejich analýzu pomocí SDS-PAGE a MALDI-TOF. Kovalentně zesítěné oligomerní formy Aβ(1-42) byly tvořeny pomocí chemické a enzymatické reakce. Prokřížení řetězců amyloidu beta vzniká přes aminokyselinový zbytek tyrosinu za vzniku kovalentní vazby, přičemž takovéto kovalentní zesítění vede ke stabilizaci oligomerní struktury, což umožňuje zlepšení analýzy oligomerů amyloidu beta. Vývoj metod pro detekci a kvantifikaci oligomerů amyloidu beta může být značným přínosem pro diagnostiku Alzheimerovy choroby, protože právě oligomerní formy amyloidu beta jsou nejvíce spojovány s patologií Alzheimerovy choroby. V teoretické části je popsána Alzheimerova choroba s uvedenými problémy v časné diagnostice a terapii. Dále jsou uvedeny základní informace o amyloidu beta a popsány analytické metody separace a detekce amyloidu beta používané ve výzkumu. Experimentální část se zabývá optimalizací analytických metod MALDI-TOF a SDS-PAGE. V metodě MALDI-TOF byly pro stanovení Aβ(1-42), jako vhodné matrice, vybrány matrice kyselina 2,5-dihydroxybenzoová a kyselina sinapová. Nejvhodnější metoda nanášení vzorku na MALDI desku pro analýzu Aβ(1-42) byla metoda vyschlé kapky. Optimalizací metody SDS-PAGE bylo zjištěno, že... This thesis focuses on the preparation of covalently cross-linked oligomeric forms of amyloid beta and their analysis by SDS-PAGE and MALDI-TOF. The covalently cross-linked oligomeric forms of Aβ(1-42) were formed by chemical and enzymatic reactions. The chains of amyloid beta are crossed over the amino acid residue of tyrosine to form a covalent bond. Such covalent cross-linking stabilizes the oligomeric structure, which allows for improved analysis of amyloid beta oligomers. The development of methods for the detection and quantification of amyloid beta oligomers may be of considerable benefit for the diagnosis of Alzheimer's disease, as it is the oligomeric forms of amyloid beta that are most strongly associated with Alzheimer's disease pathology. In the theoretical part, Alzheimer's disease is described with the above-mentioned problems in early diagnosis and therapy. In addition, background information on amyloid beta is given, and analytical methods for the separation and detection of amyloid beta used in research are described. The experimental part deals with the optimization of analytical methods MALDI-TOF and SDS-PAGE.
