[cze][eng] NK buňky hrají klíčovou roli v boji proti buňkám, které byly napadeny virem, prvokem nebo podlehly maligní transformaci. Kromě toho také regulují aktivitu a množství dalších buněk imunitního systému. Cílové buňky rozpoznávají pomocí svých aktivačních a inhibičních receptorů, ze kterých přijímají aktivační a inhibiční signály, na nichž je závislá cytotoxická odpověď NK buňky. Mezi signály panuje dynamická rovnováha, která rozhoduje o životě a smrti cílové buňky. Pokud převáží aktivační signály, bude cílová buňka eliminována. Pokud převáží inhibiční signály, pak cytotoxická odpověď nenastane. Receptor NKp30 patří do superrodiny receptorů podobných imunoglobulinům. Jedná se o důležitý aktivační receptor, který rozpoznává celou řadu ligandů, mezi které patří hemaglutinin viru vakcinie a ektromélie, protein pp65 lidského cytomegaloviru, B7-H6, BAG-6 a galektin-3. Extracelulární doména receptoru NKp30 je za určitých podmínek schopna v roztoku homooligomerizovat. První podmínkou je přítomnost N-glykosylace, druhou podmínkou je přítomnost 15 aminokyselin dlouhé "stalk" domény, která propojuje ligand vázající doménu s transmembránovým α-helixem. Cílem této práce bylo ověřit, které ze třech dostupných míst N-glykosylace receptoru NKp30 je zodpovědné za jeho homooligomerizaci. The extracellular domain of the NKp30 receptor is capable of homooligomerization in solution under certain conditions.
