[cze][eng] V teoretické části mé bakalářské práce se nejprve zaměřuji na přiblížení tématu urogenitálních infekcí. Okrajově zmiňuji i infekce, které se vyskytují u mužů, poté se věnuji především infekcím urogenitálního traktu ženy. Zmiňuji zde také sexuálně přenosné choroby. Pak přecházím přímo k tématu mé bakalářské práce, kde se věnuji blíže bakterii Streptococcus agalactiae a jejího screeningu. Praktickou část věnuji osvojením metod, které se využívají pro stanovení přítomnosti Streptococcus agalactiae. Celá praktická část byla provedena v mikrobiologické laboratoři SYNLAB s.r.o. v Českých Budějovicích. Využity byly metody běžné kultivace, kultivace, která probíhala po pomnožení patogena v Todd Hewitt Broth + Antibiotics. Pokud došlo při kultivaci ke zjištění přítomnosti Streptococcus agalactiae, musel být proveden další test, abychom mohli s jistotou říct, že se opravdu jedná o tohoto patogena. Nejčastěji dochází k dourčování pomocí MALDI-TOF MS, ale může být využita také latexová aglutinace, CAMP test či biochemická test API 20 STREP. Během výzkumu bylo zpracováno 100 vzorků, které podlehly běžné kultivaci a pak kultivaci dle doporučeného postupu, kde je výtěrový tampon ještě 24 hodin inkubován v pomnožovacím bujonu Todd Hewitt Broth + Antibiotics. Ze 100 vzorků se nám pozitivita ukázala po první kultivaci ve 14 případech, po pomnožení v bujonu bylo odhaleno ještě dalších 5. Ze získaných dat a dat poskytnutých laboratoří za 3 roky jsem zpracovala statistiku, kterou jsem dále porovnala s celorepublikovými daty. Cílem mé bakalářské práce bylo seznámení se s problematikou urogenitálních infekcí a možnostmi jejich laboratorní diagnostiky. Prováděla jsem vyšetření skupiny pacientů metodou bakteriologické kultivace výtěrů z pochvy a dále kultivaci po pomnožení v bujonu Todd Hewitt Broth + Antibiotics. Následně jsem porovnala výsledky těchto dvou metod a srovnala výsledky laboratoře s frekvencí pozitivních nálezů s celorepublikovými daty. In the theoretical part of my bachelor thesis, I first focus on the topic of genitourinary infections. I also mention infections that occur in men, then I deal mainly with infections of the genitourinary tract of a woman. I also mention sexually transmitted diseases. Then I go straight to the topic of my bachelor thesis, where I deal with Streptococcus agalactiae bacteria and its screening. The practical part is devoted to the adoption of methods that are used to determine the presence of Streptococcus agalactiae. The whole practical part was carried out in the microbiological laboratory SYNLAB s.r.o. in České Budějovice. The methods of common cultivation were used, cultivation that took place after multiplication of the pathogen in Todd Hewitt Broth + Antibiotics. If Streptococcus agalactiae was detected during cultivation, another test had to be carried out to ensure that this is indeed the pathogen. MALDI-TOF MS is the most frequent determinant, but latex agglutination, CAMP test or API 20 STREP biochemical test can also be used. During the research, 100 samples were processed, which were subjected to normal cultivation and then cultivation according to the recommended procedure, where the swab is incubated in the Todd Hewitt Broth + Antibiotics for 24 hours. Of the 100 samples, the positivity was shown to us after the first cultivation in 14 cases, after multiplication in the broth, another 5 were revealed. From the obtained data and data from the last 3 years provided by the laboratory, I compiled statistics, which I further compared with nationwide data. The aim of my bachelor thesis was to get acquainted with the issue of genitourinary infections and the possibilities of their laboratory diagnosis. I performed the examination of a group of patients by bacteriological cultivation of vaginal swabs and further cultivation after multiplication in the Todd Hewitt Broth + Antibiotics. Subsequently, I compared the results of these two methods and compared the results of the laboratory with the frequency of positive findings with the nationwide data.
