[cze][eng] Diplomová práce se zaměřuje na stanovení fyzikálně−chemických a stabilitních vlastností vybraných látek ze skupiny potenciálních antituberkulotik označovaných jako T−, K−, LV− a G− látky Cílem této práce bylo rozšířit o těchto sloučeninách portfolio informací nezbytných k výběru látek pro následné preklinické testování nových potenciálních léčiv. Teoretická část práce přináší seznámení s celosvětově rozšířeným závažným infekčním onemocněním, tuberkulózou, a vznikající rezistencí kmenů Mycobacterium tuberculosis na nynější léčiva. Dále jsou zde uvedeny vývojové směry nových potenciálních antituberkulotik a popis analytických metod použitých v experimentální části - spektrofotometrie, kapalinová chromatografie a hmotnostní spektrometrie. Experimentální část práce se věnuje popisu jednotlivých postupů, které byly použity pro stanovení vlastností vybraných látek. Jmenovitě se jedná o rozpustnost látky v 5 % DMSO, stabilitu v plasmě a Šulově půdě, mikrosomální stabilitu a experimentální stanovení logP/logD. Z výsledků experimentů je velmi dobře patrné, jak jednotlivé strukturní obměny v molekulách látek ovlivnily některé farmakokinetické či fyzikálně-chemické vlastnosti dané sloučeniny. Například snížení plasmatické stability, v důsledku začleněním síry do uhlíkového spojovacího řetězce, nebo snížení...This master's thesis focuses on determining the physio−chemical and stability properties of select groups of potential antituberculosis agents such as T, K, LV and G substances. The goal of this work was to expand the portfolio of information necessary as this for subsequent preclinical testing of new potential drugs. The theoretical work introduces the serious global infectious disease, tuberculosis, and the emerging resistance of Mycobacterium tuberculosis strains to current drugs. Furthermore, the developmental directions of new potential antituberculosis drugs and a description of the used analytical methods − spectrophotometry and HPLC analysis − are presented here. The experimental work is dedicated to the description of individual techniques which were used to determine the properties of the selected substances, such as its solubility in 5 % DMSO, stability in plasma, microsomal stability, stability in Šul media, and experimental determination of logP and logD. It is very clear from the experimental results how individual structural changes in the molecules affected some pharmacokinetic or physio−chemical properties of the given substance.
