Nádorová onemocnění jsou jednou z hlavních příčin úmrtí ve vyspělých zemích. Tradiční léčba je založena na chirurgickém odstranění nádoru, radioterapii a chemoterapii. V poslední době přibývají nové možnosti terapie, mezi které patří i imunoterapie. Imunoterapie nádorů je nepochybně velmi perspektivní variantou léčby, která v posledních několika dekádách zažila mnohé úspěchy. Nicméně stále existuje řada pacientů, která na dnešní imunoterapeutické ocnění je adoptivní buněčná imunoterapie využívající připravené imunitní buňky účastnící se boje proti Většina výzkumu se soustředí na využití T buněk, ačkoliv se pozornost upíná i na jiné buněčné typy, mezi které patří i dendritické buňky. Právě na modulaci dendritických buněk pro adoptivní buněčnou imunoterapii nádorových onemocnění se zaměřuje tato diplomová práce. Praktická část práce se věnuje hodnocení vlivu beta u na maturaci dendritických buněk monocytů. Its traditional treatment is based on surgical removal of the tumor and metastases, radiotherapy, and chemotherapy. Recently, many new therapy options, including immunotherapy, have been investigated. Cancer immunotherapy seems to be a very promising treatment option as it has experienced many successes in the last few decades. However, there is still a number of patients not responding to today's immunotherapy methods. Adoptive cellular immunotherapy is one of those immunotherapeutic methods. This immunotherapeutic modality uses ex vivo prepared immune cells that participate in anti-tumor responses. Nowadays, most research is focused on the use of T cells, although many other cell types are considered, including dendritic cells. This thesis is focused on the modulation of dendritic cells for adoptive cellular cancer immunotherapy. The aim of the practical part is to evaluate the influence of beta2-microglobulin on the maturation of monocyte-derived dendritic cells.
