[cze][eng] Nádory hlavy a krku jsou etiologicky spojené s kouřením a konzumací alkoholu. Část těchto nádorů je však iniciována HPV a jejich incidence v posledních letech významně roste. Pacienti s virově indukovanými nádory vykazují lepší prognózu, a to i přesto, že tyto nádory bývají diagnostikované v pokročilejším stádiu. Jedním z vysvětlení může být lepší stimulace imunitního systému virovými antigeny. Makrofágy jsou buňky vrozeného imunitního systému, které patří mezi profesionální fagocyty. Po infiltraci do nádoru jsou označovány jako TAM a představují heterogenní skupinu buněk, jejíž dva základní fenotypy jsou protinádorové M1 a pronádorové M2 makrofágy. U mnoha typů nádorů tvoří TAM hlavní složku nádorového mikroprostředí. Jedním z nich jsou i nádory hlavy a krku. Ve své práci jsem se zaměřila na imunohistochemickou detekci populací M1 a M2 makrofágů v nádorech hlavy a krku virové a nevirové etiologie a současně jsem metodou RT- qPCR analyzovala expresi makrofágových a/nebo imunosupresivních genů IDO1, ARG1, CD163, NOS2 a PTGS2. Ve studii jsem prokázala větší počet M2 makrofágů s typickými znaky CD163, ARG1 a PTGS2 u HPV negativních nádorů. Pacienti s těmito nádory mají horší prognózu onemocnění. Part of these tumors is induced by HPV and their incidence is increasing in the last decade. Patients with virally induced tumors have better prognosis even though they are usually diagnosed with tumors in advanced stage. One of the possible explanations may be better stimulation of the immune system by viral antigens. Macrophages are cells of the innate immune system which belong to professional phagocytes. They are called TAM upon infiltration to the tumor where they represent heterogeneous group of cells. Two main phenotypes are antitumor M1 and protumor M2 macrophages. TAMs are a major component of tumor microenvironment of many types of tumors, one of them are also head and neck cancers. In my thesis I focused on the immunohistochemical detection of M1 and M2 macrophages in the head and neck tumors of viral and non-viral etiology and at the same time RT-qPCR analyses of gene expression of macrophage-associated and/or immunosuppressive genes IDO1, ARG1, CD163, NOS2 a PTGS2 was performed. My data showed that HPV- negative tumors had higher number of M2 macrophages with typical markers CD163, ARG1 and PTGS2.
