Galectins are carbohydrate binding lectins which possess many biological activities related to the development and progression of cancer. The purpose of this bachelor's thesis is synthesis, physico- chemical and in vitro preliminary biological evaluation of novel glycopolymers with high affinity to galectin-3. Several biocompatible water-soluble linear and diblock N-(2- hydroxypropyl)methacrylamide (HPMA) copolymers bearing multivalently presented glycomimetic ligands, were synthesized and characterized by number of analytical methods, i.e. GPC, UV-VIS spectrometry, DLS and NMR. The polymer carriers were synthesized by the controlled RAFT polymerization, their molecular weight ranged from 25,000-33,000 g/mol and showed quite low dispersity. Glycopolymers containing about 4 mol% of glycomimetic ligands showed high ability to inhibit apoptosis of T lymphocytes in vitro. Nevertheless, the hypothesis consisting in the improvement of biological activity due to the denser ligand presentation on diblock copolymers was not proved. The thesis concludes that the prepared glycopolymers, linear as well as diblocks, could hypothetically be convenient tools for synergic therapy with polymer drug delivery systems in cancer treatment. Galektiny jsou lektiny vázající sacharidy, které mají mnoho biologických aktivit souvisejících s vývojem a progresí rakoviny. Cílem této bakalářské práce je syntéza, fyzikálně-chemické a předběžné in vitro biologické hodnocení nových glykopolymerů s vysokou afinitou ke galektinu- 3. Bylo syntetizováno několik biokompatibilních ve vodě rozpustných lineárních a diblokových N-(2-hydroxypropyl)methakrylamidových (HPMA) kopolymerů nesoucích multivalentně prezentované glykomimetické ligandy. Připravené glykopolymery byly charakterizovány řadou analytických metod, např. GPC, UV-VIS spektrometrií, DLS a NMR. Polymerní nosiče byly syntetizovány řízenou RAFT polymerací, jejich molekulová hmotnost se pohybovala v rozmezí 25 000-33 000 g/mol a vykazovaly poměrně nízkou disperzitu. Glykopolymery obsahující asi 4 mol % glykomimetických ligandů vykazovaly vysokou schopnost inhibovat apoptózu T- lymfocytů in vitro. Nicméně hypotéza spočívající ve zlepšení biologické aktivity díky hustší prezentaci ligandu na diblokových kopolymerech se nepotvrdila. Závěrem je možné konstatovat, že připravené glykopolymery, jak lineární, tak i diblokové, by hypoteticky mohly být vhodnými nástroji pro synergickou terapii s polymerními systémy dopravujícími léčiva pro cílenou terapii rakoviny.
