RNDr. Jakub Hofman, Ph.D. Název diplomové práce: Studium interakcí vybraných cílených léčiv s ABC lékovými transportéry Nádorová onemocnění patří mezi jednu z nejčastějších příčin úmrtí a jejich výskyt se neustále zvyšuje. Léčba zahrnuje především kombinaci medikamentózní terapie s chirurgickou léčbou a radioterapií. Jedním z novějších přístupů s obrovským potenciálem je použití tzv. cílené protinádorové léčby, která specificky interaguje s nádorovými buňkami, čímž omezuje negativní dopady klasické chemoterapie. I přes nepřetržitý intenzivní výzkum nových přístupů v terapii se stále potýkáme s nedostatečnou odpovědí na léčbu. Hlavní příčinou selhání terapie je zejména vznik mnohočetné lékové rezistence (MDR) a tvorba metastáz. MDR fenomén souvisí nejčastěji s nadměrnou expresí ATP binding cassette (ABC) transportérů, které jsou zodpovědné nejen za nadměrný eflux protinádorových léčiv z buněk, ale také zásadním způsobem ovlivňují jejich farmakokinetiku. Inhibicí těchto transportérů dochází ke zvýšení intracelulární koncentrace cytostatika a možnosti modulovat MDR. Treatment mainly includes a combination of drug therapy, surgery and radiotherapy. A new approach, with huge potential, is the use of targeted anti-tumor therapy. This approach is based on specific interaction with tumor cells that reduces the negative effects occuring in conventional chemotherapy. Despite intensive research, we still face insufficient response to treatment. The main cause of therapy failure is the emergence of multidrug resistance (MDR) and the formation of metastases. The MDR phenomenon is most often associated with the overexpression of ATP binding cassette (ABC) transporters. These are responsible not only for the excessive efflux of anticancer drugs from cells, but also mainly affect their pharmacokinetics. Inhibition of these transporters increases the intracellular concentration of the cytostatic and provide the ability to modulate MDR.
