RNDr. Lenka Skálová, Ph.D. Název diplomové práce: Účinek thujonu a piperitonu na expresi biotransformačních enzymů v jaterních řezech Monoterpeny jsou přírodní látky, které jsou široce rozšířené v silicích celé řady druhů rostlin. Thujon a piperiton jsou obsaženy v některých druzích rostlin a mohou být součástí potravinových doplňků a jiných rostlinných přípravků nebo potravin, kde slouží jako dochucovadla. Cílem této diplomové práce bylo zjistit, jak thujon a piperiton ovlivňují expresi vybraných biotransformačních enzymů z nadrodiny cytochromů P450, sulfotransferas a UDP-glukuronosyltransferas. Pro měření byly použity ultratenké jaterní řezy získané z jater 3 lidských dárců. Řezy byly inkubovány s thujonem nebo piperitonem v koncentracích 10 μM a 50 μM po dobu 24 hodin. Po inkubaci bylo kvantifikováno množství mRNA a proteinů vybraných enzymů. Získané výsledky vykazovaly značnou interindividuální variabilitu. Exprese CYP1A2 na úrovni mRNA byla signifikantně zvýšena po ovlivnění řezů thujonem (50 μM) a piperitonem (10 μM i 50 μM), avšak pouze u jednoho dárce. Účinkem piperitonu (50 μM) byla zvýšena mRNA pro UGT1A9 rovněž pouze u jednoho dárce. Zvýšení množství mRNA... Thujone and piperitone are found in certain plant species that can be part of dietary supplements, herbal preparations or used in foods as flavourings. The aim of this diploma thesis was to find out how thujone and piperitone affect the expression of selected biotransformation enzymes from the cytochrome P450 superfamily, the sulfotransferase and the UDP-glucuronosyltransferase superfamilies. Precision-cut liver slices obtained from the livers of 3 human donors were used for these experiments. Slices were incubated with thujone or piperitone for 24 hours at 10 μM and 50 μM concentrations. After incubation, the amount of mRNA and protein was quantified. The obtained results showed considerable interindividual variability.
