Histony a jejich N a C konce podléhají kovalentním modifikacím, které regulují transkripci genů. Mezi histonové modifikace patří methylace, ubikvitinylace, SUMOylace, ADP- ribosylace, fosforylace, prolinová izomerace, deiminace a acetylace. Acetylace histonů je regulována histonacetyltransferasami (HATs) a histondeacetylasami (HDACs). Rovnováha mezi acetylací/deacetylací histonů ovlivňuje kompaktnost chromatinu a podílí se na regulaci genové exprese. U řady lidských nádorů je acetylační rovnováha porušena a může přispívat k rozvoji maligních onemocnění. Inhibitory histondeacetylas (HDACi) umožňují tuto porušenou acetylační rovnováhu vyrovnávat. Jeden z těchto inhibitorů je kyselina valproová (VPA), již používaná k léčbě epilepsie. V několika studiích bylo prokázáno, že VPA vykazuje protinádorové účinky. U nádorových neuroblastomových linií byla zaznamenána např. snížená exprese n-myc onkoproteinu, inhibice buněčného růstu či angiogeneze. Ačkoliv je přesný mechanizmus účinku VPA na nádorové buněčné linie stále nejasný, za jeden z důležitých protinádorových účinků je povážována hyperacetylace histonů H3 a H4. V této práci je ukázáno, že kyselina valproová zvyšuje acetylaci histonů H3 a H4 v lidských neuroblastomových buněčných liniích UKF-NB-3 a SK-N-AS. Nejvyšší acetylace histonu H3 byla v hypoxii... Histones and their N and C terminal tails undergo different covalent modifications that regulate gene transcription. Among these histone modifications are methylation, ubiquitinilation, SUMOylation, ADP- ribosylation, phosphorylation, proline izomerization, deimination and acetylation. Histone acetylation is regulated by histonacetyltransferases (HATs) and histondeacetylases (HDACs). The balance between acetylation/deacetylation influences chromatin condensation and thus regulates gene transcription. Acetylation balance is disrupted in many human cancers and this fact can contribute to the development of malignant diseases. Histondeacetylase inhibitors (HDACi) can restore this acetylation imbalance. One of these HDACi is valproic acid (VPA) which has been used in treatment of epilepsy for decades. VPA shows antitumour effect in many studies. Decreased expression of n-myc oncoprotein, inhibition of tumour growth and angiogenesis are one of these anticancer effects observed in neuroblastoma cell lines after treatment with VPA. Despite the fact that exact mechanism of antitumour effect of VPA remains unclear, one of the most important mechanisms is hyperacetylation of histone H3 and H4.
