[cze][eng] Chřipkový virus v posledních letech vzbudil hned několikrát celosvětové obavy ze vzniku pandemie. Nejzávažnějším problémem v boji proti tomuto viru je jeho schopnost neobvykle rychle měnit antigenní specifitu jednak přeskupením genových segmentů, jednak pomocí bodových mutací, a to především v povrchových glykoproteinech - hemaglutininu a neuraminidáze. V této práci jsem se zaměřila na bodové mutace zasahující hemaglutinin chřipkového viru, které způsobují změnu vazebné preference. Tato změna následně umožňuje chřipkovému viru jednak přenos mezi různými hostiteli a také neomezené šíření v jejich tkáních. Neustálá změna aminokyselinové sekvence a tím i povrchu hemaglutininu také do značné míry znemožňuje imunitnímu systému hostitele virus efektivně zlikvidovat a dostatečně rychle rozpoznat při další nákaze. Každý ze subtypů hemaglutininu má ovšem jiné pozice aminokyselin ovlivňující jeho vazebnou preferenci. Jejich souhrn je obsažen v této práci. The most serious problem, in the fight against this virus, is its ability to quickly change its unusually antigenic specificity by reassortment and point mutations, especially in surface glycoproteins - haemagglutinin and neuraminidase. In this thesis I focused on point mutations in the hemagglutinin of influenza virus causing change in binding preference, which enables both influenza virus transmission between different hosts and unlimited proliferation in their tissues. The permanent change in amino acid sequence and thus the surface of hemagglutinin also largely prevents the host's immune system to destroy the virus efficiently and quickly enough to recognize when another infection. Each of the haemagglutinin subtypes has a different amino acid positions affecting its binding preference. Their summary is included in this thesis.
