[cze][eng] 1 Souhrn V předložené rigorózní práci jsou podrobně popsány různé metody přípravy farmaceutických nanočástic, jejich povrchové vlastnosti a uvolňování v nich obsažených léčiv. Jsou prezentovány některé důležité informace týkající se perspektivních možností nanosystémů ve farmakoterapii. Práce je především zaměřena na experiment. Experimenty vyúsťují do využití zařízení na měření velikosti nanočástic a jejich zeta potenciálu. Byly připraveny nanočástice z různých oligoesterů s lineární a větvenou konstitucí řetězce menší než 200 nm. Hlavní záměr předložené práce je soustředěn na stálost parametrů velikosti částic a jejich povrchového náboje při jejich uchovávání při různých teplotních podmínkách a při statických a dynamických podmínkách ve vnější fázi po přípravě nanočástic. Kapalné systémy s nanočásticemi byly těsně po přípravě sprejové sušeny s mannitolem jako aditivem. Byly zkoumány procesy, které obsahovaly řadu stresových faktorů, jako je rozpouštění mannitolu jako osmotického agens, operace sprejového sušení, dispergace nanočástic obsažených v mikročásticích ve vodném médiu. Tyto procesy byly hodnoceny z hlediska změn velikosti a zeta potenciálu nanočástic. Došlo se k závěru, že metoda založená na sprejovém sušení nanosystémů s inkorporací nanočástic je výhodná a užitečná při stabilizaci těchto nanosystémů.1 Summary In the thesis are in details described various methods of pharmaceutical nanoparticles preparation, their surface properties and incorporated drug release. Some relevant data concerning prospective use of nanosystems in pharmacotherapy are presented. The main orientation of the thesis is towards experimental laboratory working. Experiments are supported by the use of Nanosizer and Zetasizer device. Nanoparticles with the size under 200 nm from various oligoesters with the linear and branched constitution of carrier molecules were fabricated. The aim of the thesis was focused on stability of size and surface charge parameters during their storage under different thermal conditions and under static and dynamic state of hydrophilic external phase after preparation process. Liquid systems containing freshly prepared nanoparticles were spray dried with the mannitol used as additive. The processes including range of stress factors, such as mannitol as osmotic agent dissolution, spray drying operation, and nanoparticles contained in microparticles in aqueous medium dispersion were evaluated from the point of view of size and zeta potential parameters.
