U některých seskviterpenů byl prokázán protinádorový účinek. Doxorubicin je antracyklinové chemoterapeutikum využívané mimo jiné v léčbě karcinomu prsu. Terapie doxorubicinem je limitována mnohými nežádoucími účinky, především pak jeho kardiotoxicitou. Kombinací doxorubicinu s vhodnými látkami by se mohlo docílit nižších terapeutických dávek doxorubicinu. Jednou z alternativ je využití seskviterpenů v kombinaci s doxorubicinem. Cílem této práce je stanovení vlivu dvou seskviterpenů α-humulenu a trans-nerolidolu na buněčnou linii karcinomu prsu MCF-7 a určení vlivu α-humulenu a trans-nerolidolu na účinek cytostatika doxorubicinu. Vliv α-humulenu, trans-nerolidolu, doxorubicinu a jejich kombinace na proliferaci buněk byl hodnocen pomocí testu neutrální červeně. Výsledky ukázaly antiproliferační účinek vyšších koncentrací α-humulenu i trans-nerolidolu na buněčnou linii MCF-7, a to po 24 hodinovém i 48 hodinovém ovlivnění. Při koncentraci... Anti-tumour effect of certain sesquiterpenes has been determined. Doxorubicin is an antracycline- based medicine used for cancer chemotherapy, among other things, for the treatment of breast cancer. The doxorubicin therapy is limited by a number of adverse effects, particularly its cardiotoxicity. A combination of doxorubicin with suitable substances might result in lower therapeutic dosages of doxorubicin. One of the alternatives is the utilization of sesquiterpenes combined with doxorubicin. The purpose of this thesis is the determination of the effect of two sesquiterpenes, α-humulene and trans-nerolidol on the MCF-7 breast cancer cell line and the determination of the effect of α-humulene and trans-nerolidol on the effect of the doxorubicin cytostatic.
