Cilem teto rigorozni prace bylo detekovat změny endotelialni exprese nitrotyrosinu ve stěně aortalniho oblouku kraliků, kterym bylo podavano antracyklinove cytostatikum daunorubicin. Sledovali jsme připadne změny exprese nitrotyrosinu po různych davkovych intervalech podavani daunorubicinu. Chronicka antracyklinova kardiotoxicita byla navozena opakovanym podanim daunorubicinu (3 mg/kg=50 mg/m2 i.v., 1x tydně) po dobu 10 tydnů. Pro sledovani časoveho morfologickych, změn aorty v průběhu rozvoje toxickeho učinku antracyklinů bylo utvořeno 10 daunorubicinovych skupin (n=6), ktere reprezentovaly zviřata exponovana různe kumulativni davce DAU. Zviřata byla ukončovana za 24 hodin nebo za 7 dnů po dane kumulativni davce cytostatika. Vysledky byly porovnany s hodnotami naměřenymi u kontrolnich zviřat (n=6), kterym nebyla aplikovana žadna latka. Imunohistochemicka analyza neprokazala indukci endotelialni exprese markeru nitračniho stresu nitrotyrosinu ani u kontrolni a ani u žadne z daunorubicinovych skupin. Lze tedy konstatovat, že daunorubicin zřejmě neindukuje morfologicky detekovatelnou endotelialni dysfunkci v aortalnim oblouku kralika u modelu antracyklinove cardiotoxicity a ukazuje na větši obrannou schopnost endotelialnich buněk ve srovnani s kardiomyocyty. 6 The aim of this thesis was to evaluate possible changes of nitrotyrosine expression in rabbit aorta after repeated administration of daunorubicin. Chronic anthracycline cardiotoxicity was induced by repeated administration of daunorubicinu (3 mg/kg=50 mg/m2 i.v., 1x week) for the period of 10 weeks. We focused on the monitoring nitrotyrosine expression in rabbit aorta. Ten daunorubicin groups were compared with control rabbits. The animals were killed 24 hours and/or 7 days after the administration of daunorubicin. Immunohistochemical analysis showed no expression of nitrotyrosine in any control or experimental groups. In conclusion, the administration of daunorubicin did not induce nitrotyrosine expression in aorta in either control and daunorubicin treated rabbits, suggesting that endothelial dysfunction and/or nitrative stress in aorta is not triggered by daunorubicin treatment in vivo. 7
