PharmDr. Tomáš Šimůnek, Ph.D. Školitel - konzultant: RNDr. Petra Špidlová, Ph.D. Název diplomové práce: Komplementace dsbA mutantního kmene Francisella tularensis a analýza potenciálních vazebných partnerů proteinu DsbA Konzervovaný hypotetický lipoprotein FTT1103 je virulenčním faktorem gramnegativní intracelulární bakterie F. tularensis. Tento protein je homologní k proteinům disulfid oxidoreduktázové rodiny DsbA. Proteiny této rodiny katalyzují tvorbu disulfidických vazeb, které jsou nezbytné pro utvoření správné konformace proteinů, pro jejich aktivitu a stabilitu. Cílem práce byla identifikace vazebných partnerů lipoproteinu FTT1103 běžně nazývaného DsbA, jejichž znalost by nám mohla pomoci v hlubším pochopení role lipoproteinu DsbA ve virulenci F. tularensis. Pomocí metod molekulární biologie jsme vytvořili fúzní gen ftt1103 se sekvencí kódující FLAG® kotvu a klonovali ho do plazmidového vektoru replikujícího se ve F. tularensis. Plazmidový vektor jsme elektroporací vnesli do in frame delečního mutanta F. tularensis subsp. holarctica kmen FSC200/∆1103, čímž došlo k jeho komplementaci in trans. This protein shares homology with proteins of disulfide oxidoreductase DsbA family. These proteins catalyse formation of disulfide bonds, which are essential for forming proteins conformation, for activity and stability of proteins. The aim of this study was to identify binding partners of lipoprotein FTT1103, commonly known as DsbA. The knowledge of these partners could help us in understanding a role of lipoprotein DsbA in virulence F. tularensis. We prepared fused ftt1103 gene with sequence coding FLAG® tag and cloned this fused gene into plasmid vector which can be replicated in F. tularensis.
