Nedávné výzkumy ukazují na přítomnost minoritní frakce nádorových buněk, která vykazuje určité "kmenové" vlastnosti. Tyto tzv. rakovinné kmenové buňky (RKB) se vyznačují relativně vyšší odolností k léčbě, tradičně zaměřené na diferencované a rychle se dělící buňky a proto mohou být příčinou relapsů nádorových onemocnění. RKB sdílejí specifickou expresi genů a deregulaci signálních drah. Jednou ze známých deregulací je zvýšená exprese receptoru erbB-2 (HER2), vyskytující se u ~25-30% případů rakoviny prsu. Zvýšená exprese erbB-2 receptoru je spojená se špatnou prognózou, zvýšeným proliferačním potenciálem a s rezistencí vůči chemoterapii. Dle nedávných výzkumů se při zvýšení hladiny erbB-2 zvyšuje i procento RKB v populaci nádorových buněk. V této diplomové práci jsme jako model RKB rakoviny prsu in vitro studovali tzv. mammosféry, rostoucí v médiu bez obsahu séra, do něhož jsou buňky postupně převáděny. Tyto buňky jsou CD44+ , vyznačují se zvýšenou expresí ABCG-2, Sox-2, Vimentinu a také vysokými hladinami erbB-2. Dále jsme připravili stabilní linii MCF7 buněk se sníženou hladinou erbB-2 pomocí shRNA plazmidu. V erbB-2- buňkách jsme charakterizovali expresi sady markerů RKB a sledovali schopnost těchto buněk iniciovat tvorbu nádoru v nahé myši pomocí ultrazvuku. Klíčová slova Rakovinné kmenové... Recent studies indicate the existence of a subpopulation of cells within tumours with stem cell-like characteristics. These "cancer stem-like cells" (CSCs) are relatively resistant to established therapies, usually targeting differentiated and fast proliferating cells. Therefore, CSCs may be a reason for the relapse of neoplastic diseases. CSCs can be characterised by a specific gene expression profile and deregulated signalling pathways. Of these, upregulation of the erbB-2 (HER2) receptor, a hallmark of ~25-30% breast cancer patients, is related to dismal prognosis, elevated proliferation potential and resistance to chemotherapy. Recent evidence has suggested that upregulation of erbB-2 leads to increase in the pool of CSCs. In our study we used mammospheres, cells grown in the absence of serum, an in vitro model of breast CSCs, which were prepared by "weaning" breast cancer MCF7 cells to a special medium. These cells were CD44high and showed increased expression of ABCG-2, Sox-2, Vimentin as well as high levels of erbB-2. Next, we prepared a stable line of MCF7 cells with low levels of erbB-2 by shRNA.
