Polyomaviry jsou malé DNA viry, které jsou schopné indukovat širokou škálu nádorů. U myšího polyomaviru (MPyV) se v indukci nádorové transformace uplatňuje zejména střední T antigen (MT antigen). MT antigen nemá vlastní enzymatickou aktivitu. Jeho funkce spočívá v aktivaci drah signální transdukce buňky, díky jeho schopnosti interagovat s určitými buněčnými proteiny. K těmto proteinům patří například protein fosfatáza 2A (PP2A), Src kináza, fosfatidyl inositol 3 kináza (PI3K), protein Shc, 14-3-3 protein nebo fosfolipáza Cγ1 (PLCγ1). Tato práce je zaměřena na interakci MT antigenu s těmito buněčnými proteiny a na dopad této interakce na transformaci buněk. Protože je MT antigen silným onkogenem, práce se zabývá i charakterem transformovaných buněk a vývojem nádoru v epitelu mléčné žlázy myši. Klíčová slova: polyomaviry, MT antigen, PP2A, PI3K, PLCγ1, protein Shc, protein 14-3-3 protein Polyomaviruses are small DNA viruses, which are able to induce a broad variety of tumors. The main oncoprotein of the mouse polyomavirus (MPyV) is middle T antigen (MT antigen) which is able to transform cells. MT antigen has not an enzymatic activity of its own. It is able to activate signal transduction of host cells through its interactions with certain cellular proteins. These proteins include protein phosphatase 2A (PP2A), Src kinase, phosphatidylinositol 3 kinase (PI3K), Shc protein, 14-3-3 protein and phospholipase Cγ1 (PLCγ1). This work is focused on interaction between MT antigen and cellular proteins and on the impact of this interaction on cell transformation. Since MT antigen is a potent oncogene, the work also deals with the character of transformed cells and tumor development in mouse mammary epithelium.
